Immortalization of Human B Cells: Analysis of B Cell Repertoire and Production of Human Monoclonal Antibodies
One of the major challenges in human B cell immunology field has been the objective to establish stable monoclonal cells lines that express the B cell receptor (BCR) on their cell surface and secrete antibodies. Such a system is extremely attractive not only for studying various aspect of BCR signaling but also for the generation of human monoclonal antibody and the analysis of the human B cell repertoire. This chapter describes an efficient method to immortalize and clone human B cells by Epstein–Barr Virus (EBV) transformation.

Causes Of Difficulty Swallowing And Swelling Of The Throat Identified By Food Elimination Diet
A six-food elimination diet significantly improves symptoms in adult patients with eosinophilic esophagitis (EoE), according to a new study in Gastroenterology, the official journal of the American Gastroenterological Association. In EoE, eosinophils and other inflammatory cells cause inflammation of the esophagus in response to an allergic stimulus. Previously thought to be a rare disease, EoE has become one of the most common causes for dysphagia (difficulty swallowing), heartburn and the sensation of "food stuck in the throat" in adults...

Study published showing how easily pandemic H5N1 bird flu could evolve
Scientists discovered it took just five genetic mutations for a potentially pandemic strain of H5N1 bird flu virus to emergeResults from a controversial research project in which scientists created a pandemic strain of the deadly H5N1 bird flu virus in the lab were finally published on Thursday, after an eight-month delay while scientists and biosecurity officials debated how much of the information should be made public.Scientists wanted to know what changes in the H5N1 virus, which cannot currently spread between people, were necessary for it to become transmissible through the air via coughs and sneezes. They geneticall...

Immune system molecule weaves cobweb-like nanonets to snag intestinal microbes
A multidisciplinary team of researchers has found that human alpha-defensin 6 (HD6) — a key component of the body's innate defense system — binds to microbial surfaces and forms "nanonets" that surround, entangle and disable microbes, preventing bacteria from attaching to or invading intestinal cells.
UCLA professor emeritus of medicine Dr. Robert I. Lehrer, whose laboratory was the first to discover defensins in the early 1980s, was co-senior author of the research, which was led by scientists from the UC Davis Health System.
The research describes an entirely new mechanism of action for defensin...

Allergies Become Epidemic: Food Allergy Sufferers Double In The Last 10 Years
This week the European Academy of Allergy and Clinical Immunology (EAACI) launched its Food Allergy Campaign. The purpose of the campaign is to raise awareness of the sharp increase of anaphylaxis in children, an allergic reaction that is severe and potentially life-threatening. It aims at educating the public to recognise the symptoms and its triggers, and to teach methods of how to react in case of emergency, e.g. by using an adrenaline pen...

Drug combo much better than AZT alone at preventing mother-to-infant HIV transmission
Non-breastfed babies born to HIV-positive mothers who didn't receive antiretroviral therapy during pregnancy are routinely given zidovudine, commonly known as AZT, shortly after birth to prevent mother-to-child transmission of the virus that causes AIDS.
While effective, this strategy doesn't always protect the infant from acquiring the virus during the mother's labor and delivery. But a new UCLA-led study published June 21 in the New England Journal of Medicine finds that a two- or three-drug combination given to infants within 48 hours of birth can reduce the risk of such intrapartum HIV acquisition by about half,...

Malaria's Spread May Be Encouraged By Rising Type 2 Diabetes Prevalence
The mosquito's immune system is suppressed by human insulin, which could be bad news for the spread of malaria if the number of people in Africa with diabetes type 2 continues to grow at its present rate, researchers from the University of California, Davis, explained in the journal Infection and Immunity. The authors added that the malaria parasite spread is slowed down by the mosquitoes' immunological resistance. First author, Nazzy Pakpour, said "A fair portion actually fight off the infection." Type 2 diabetes rates in Africa, as in the rest of the world, are rising rapidly...

Working Safely with Vaccinia Virus: Laboratory Technique and Review of Published Cases of Accidental Laboratory Infections
Vaccinia virus (VACV), the prototype orthopoxvirus, is widely used in the laboratory as a model system to study various aspects of viral biology and virus–host interactions, as a protein expression system, as a vaccine vector, and as an oncolytic agent. The ubiquitous use of VACVs in the laboratory raises certain safety concerns because the virus can be a pathogen in individuals with immunological and dermatological abnormalities, and on occasion can cause serious problems in normal hosts. This chapter reviews standard operating procedures when working with VACV and reviews published cases on accidental laboratory infections.

Analyzing CD8 T Cells in Mouse Models of Poxvirus Infection
Mouse models of immunology are frequently used to study host responses to poxviruses or poxvirus-based recombinant vaccines. In this context, the magnitude of CD8+ T cell responses is often of interest. Methods to evaluate CD8+ T cell responses extend from those that rely on indirect measurement of effector function only, such as cytotoxicity assays, to those that only measure antiviral CD8+ T cell numbers and not function, like peptide MHC tetramers. In this chapter, five methods are provided that cover this range: DimerX staining (a variant of peptide-MHC tetramers), intracellular cytokine staining for interferon-γ...

Meta-analysis: Vitamin D plus calcium and effect on mortality
Source: J Clin Endocrinol Metab
Area: News
Cross-sectional and longitudinal studies have linked reduced vitamin D status with a number of cancers, adverse cardiovascular and immunological outcomes, and increased all-cause mortality. A meta-analysis of randomized controlled trials of vitamin D supplementation published in 2007 found that vitamin D reduced all-cause mortality by 7%, however, three subsequent meta- failed to show a statistically significant effect of vitamin D alone on mortality, although two of the studies did show a reduced mortality if vitamin D was given together with calcium. This meta-analysis of indi...
