On the selectivity of neuronal NOS inhibitors
Conclusion and ImplicationsThe results suggest that l‐VNIO, NPA and 1400W, as well as the newer pyrrolidine‐type inhibitors, cannot be used as neuronal NOS inhibitors in cells without stringent verification. The identification of inhibitors with useable selectivity in cells and tissues remains an important goal.

The inhibitor of volume‐regulated anion channels DCPIB activates TREK potassium channels in cultured astrocytes
Conclusion and ImplicationsOur results indicate that the neuroprotective effect of DCPIB could be due, at least in part, to activation of TREK channels. DCPIB could be used as template to build new pharmacological tools able to increase background K+ conductance in astroglia and neuronal cells.

Noradrenaline stimulates cell proliferation by suppressing potassium channels via Gi/o‐protein‐coupled α1B‐adrenoceptors in human osteoblasts
Conclusions and ImplicationsNoradrenaline facilitated cell proliferation by regulation of potassium currents in human osteoblasts via Gi/o‐protein‐coupled α1B‐adrenoceptors, not via coupling to Gq‐proteins.

The anti‐cancer agent SU4312 unexpectedly protects against MPP+‐induced neurotoxicity via selective and direct inhibition of neuronal NOS
Conclusions and ImplicationSU4312 exhibited neuroprotection against MPP+ at least partly via selective and direct inhibition of nNOS. Because SU4312 could reach the brain in rats, our study also offered a support for further development of SU4312 to treat neurodegenerative disorders, particularly those associated with NO‐mediated neurotoxicity.

Anti‐inflammatory effects of tobramycin and a copper–tobramycin complex with superoxide dismutase‐like activity
Conclusions and ImplicationsAnti‐inflammatory effects of tobramycin in vivo may relate to the spontaneous formation of a copper–tobramycin complex, implying that copper–tobramycin may be more effective therapy.

Effects of μ‐opioid receptor modulation on the hippocampal network activity of sharp wave and ripples
Conclusions and ImplicationsWe found that the SWRs were significantly modulated by three MOR agonists and that the SWRs were very sensitive to subtle changes in the excitation/inhibition balance induced by MOR agonists. Such modulation might underlie the effects of these agonists on hippocampus‐dependent memory.

4‐O‐methylhonokiol, a PPARγ agonist, inhibits prostate tumour growth: p21‐mediated suppression of NF‐κB activity
Conclusions and ImplicationMH inhibits growth of human prostate cancer cells through activation of PPARγ, suppression of NF‐κB and arrest of the cell cycle. Thus, MH might be a useful tool for treatment of prostate cancer.

A study of subunit selectivity, mechanism and site of action of the delta selective compound 2 (DS2) at human recombinant and rodent native GABAA receptors
Conclusions and ImplicationsDS2 is the first PAM selective for α4/6βxδ receptors, providing a novel tool to investigate extrasynaptic δ‐GABAA receptors. The effects of DS2 are mediated by an unknown site leading to GABAA receptor isoform selectivity.

ONO‐2506 inhibits spike–wave discharges in a genetic animal model without affecting traditional convulsive tests via gliotransmission regulation
Conclusions and ImplicationsONO‐2506 did not affect traditional convulsive tests but markedly inhibited epileptic phenomena in the genetic epilepsy mouse model. ONO‐2506 enhanced release of inhibitory neuro‐ and gliotransmitters during the resting stage and inhibited tripartite transmission during the hyperactive stage. The results suggest that ONO‐2506 is a novel potential glial‐targeting antiepileptic drug. Linked ArticleThis article is commented on by Onat, pp. 1086–1087 of this issue. PPAR‐γ – a possible drug target for complicated pregnancies
Peroxisome proliferator activated receptors (PPARs) are ligand‐activated transcription factors expressed in trophoblasts, which regulate both cell differentiation and proliferation. In recent years, evidence has linked PPARs to playing an integral role in pregnancy; specifically, PPAR‐β and PPAR‐γ have been shown to play an integral role in placentation, with PPAR‐γ additionally serving to regulate trophoblast differentiation. Recent evidence has shown that PPAR‐γ expression is altered in many complications of pregnancy such as intrauterine growth restriction (IUGR), preterm birth, pre‐clampsia and gestatio...

IFN‐α subtypes: distinct biological activities in anti‐viral therapy
During most viral infections, the immediate host response is characterized by an induction of type I IFN. These cytokines have various biological activities, including anti‐viral, anti‐proliferative and immunomodulatory effects. After induction, they bind to their IFN‐α/β receptor, which leads to downstream signalling resulting in the expression of numerous different IFN‐stimulated genes. These genes encode anti‐viral proteins that directly inhibit viral replication as well as modulate immune function. Thus, the induction of type I IFN is a very powerful tool for the host to fight virus infections. Many viruses...

Cohort study: Hyperbaric oxygen therapy not effective for diabetic foot ulcers and the prevention of amputation
Source: Diabetes Care Area: News A cohort study has found that hyperbaric oxygen (HBO) did not improve wound healing or prevent lower extremity amputation in patients with diabetic foot ulceration   The researchers studied 6,259 individuals representing 767,060 person-days of wound care. In propensity score-adjusted models, individuals receiving HBO were less likely to have healing of their foot ulcer (hazard ratio, 0.68; 95% confidence interval, 0.63-0.73) and more likely to have an amputation (2.37 [1.84-3.04]) than those patients who did not receive HBO.   The authors concluded that the usefulness of HB...

RCT: Bendamustine plus rituximab vs CHOP plus rituximab as first-line treatment for indolent and mantle-cell lymphomas
Source: Lancet Area: News Rituximab plus chemotherapy, most often CHOP (cyclophosphamide, doxorubicin, vincristine, and prednisone), is considered the first-line standard of care for patients with advanced indolent lymphoma, and for elderly patients with mantle-cell lymphoma, however there are no randomised comparative study data to show that this regimen is better than other chemotherapy regimens. Bendamustine plus rituximab is effective for relapsed or refractory disease and based on long-standing experience of its use in Germany, this prospective, randomised, open-label non-inferiority study compared this regimen with ...

Meta-analysis: Hydroxyethyl starch associated with mortality and acute kidney injury in critically ill patients
Source: JAMA Area: News Hydroxyethyl starches are synthetic fluid products widely used for volume resuscitation but clinical trial data are conflicting and have suggested an increased risk of mortality and acute kidney injury. Furthermore, multiple trials from one researcher have been retracted because of scientific misconduct.   This systematic review and meta-analysis evaluated the association of hydroxyethyl starch on these outcomes with an additional investigation on the influence of the retracted studies.   Investigators searched various databases (to October 2012), reference lists of relevant artic...
