Latest research and development trends in non-insulin anti-diabetics.
Abstract Type 2 diabetes mellitus, also called non-insulin dependent diabetes mellitus, is a chronic endocrine disease characterized by insulin resistance in tissues such as fat, liver and skeletal muscle, and impaired insulin secretion in pancreatic β cells. The prevalence and incidence of type 2 diabetes exploded over last decades along with increased population obesity owing to western lifestyle factors such as lack of exercise and high calorie diets. As diabetes progresses without appropriate treatment, many micro- and macro-vascular complications occur, leading to increased risk of mortality. Although lifesty...

Administration of a Loading Dose of Atorvastatin Before Percutaneous Coronary Intervention Prevents Inflammation and Reduces Myocardial Injury in STEMI Patients: A Randomized Clinical Study.
CONCLUSIONS: Loading-dose atorvastatin therapy before emergency PCI reduced the inflammatory response and myocardial dysfunction in these STEMI patients by lowering hs-CRP, BNP, and MMP-9. Pre-PCI loading-dose atorvastatin treatment may help prevent inflammatory response and improve cardiac function in patients with acute coronary syndromes undergoing emergency PCI. Sirtuins as Possible Targets in Neurodegenerative Diseases.
Abstract Age-related diseases pose as an enormous problem on aging populations of the world. Despite the fact that many advances have been made on understanding of the neurodegeneration, there is still no cure available for the age-related brain disorders. Sirtuins are NAD-dependent protein deacetylases that were shown to have beneficial effects against age-related diseases. SIRT1 and SIRT2 have been studied mostly in terms of neurodegenerative diseases and seem to have opposite effects. According to the recent findings, activators of SIRT1 and inhibitors of SIRT2 would benefit the brain from neurodegeneration. Des...

The Use of Keratin in Biomedical Applications.
Abstract Keratins are naturally derived proteins that can be fabricated into several biomaterials morphologies including films, sponges and hydrogels. As a physical matrix, keratin biomaterials have several advantages of both natural and synthetic materials that are useful in tissue engineering and controlled released applications. Like other naturally derived protein biomaterials, such as collagen, keratin possess amino acid sequences, similar to the ones found on extracellular matrix (ECM), that may interact with integrins showing their ability to support cellular attachment, proliferation and migration. The abil...

Identification of Novel Drug Targets in Hpb38, Hpp12, Hpg27, Hpshi470, Hpsjm180 Strains of Helicobacter Pylori: An In Silico Approach for Therapeutic Intervention.
Abstract Helicobacter species colonizes the stomach and are associated with the development of gastritis disease. Drugs for treatment of Helicobacter infection relieve pain or gastritis symptoms but they are not targeted specifically to Helicobacter pylori. Therefore, there is dire need for discovery of new drug targets and drugs for the treatment of H. pylori. The main objective of this study is to screen the potential drug targets by in silico analysis for the potent strains of H. pylori which include HpB38, HpP12, HpG27, Hpshi470 and HpSJM180. Genome and metabolic pathways of pathogen H. pylori and the host Homo...

Integration of Gene Expression and ChIP-chip Analyses Uncovers Genome-Wide Androgen Receptor Signaling in Prostate Cancer.
Abstract The androgen receptor (AR) is a steroid hormone receptor that functions as a ligand-dependent transcriptional factor, which plays a key role in the development and progression of prostate cancer. Recent advance in high-throughput technologies including microarrays and deep-sequencing provides unbiased genome-wide knowledge on the AR signaling including datasets for androgen-regulated gene expression and genomic binding sites for AR. In the present review, we will briefly summarize the main features of the AR signaling as well as the individual AR target genes identified by the integration of multiple datas...

Androgen Receptor Gene Rearrangements: New Perspectives on Prostate Cancer Progression.
Abstract The androgen receptor (AR) is a master regulator transcription factor in normal and cancerous prostate cells. Canonical AR activation requires binding of androgen ligand to the AR ligand binding domain, translocation to the nucleus, and transcriptional activation of AR target genes. This regulatory axis is targeted for systemic therapy of advanced prostate cancer. However, a new paradigm for AR activation in castration-resistant prostate cancer (CRPC) has emerged wherein alternative splicing of AR mRNA promotes synthesis of constitutively active AR variants that lack the AR ligand binding domain (LBD). Rec...

The Role of Tumor Suppressor Dysregulation in Prostate Cancer Progression.
Abstract Androgen receptor activity is essential for prostate cancer development and progression. While there are classically defined roles for the retinoblastoma (Rb) and p53 tumor suppressor pathways in maintenance of cell cycle control and the DNA damage response, recent studies have demonstrated a direct role of these two pathways in regulating AR expression and function. While the role of Pten deregulation in prostate cancer has provided much insight in to the mechanisms underlying prostate cancer initiation and progression, emerging roles for Rb and p53 are likely to further expand upon our understanding of t...

Recurrent Rearrangements in Prostate Cancer: Causes and Therapeutic Potential.
Abstract DNA damage and genetic rearrangements are hallmarks of cancer. However, gene fusions as driver mutations in cancer have classically been a distinction in leukemia and other rare instances until recently with the discovery of gene fusion events occurring in 50 to 75% of prostate cancer patients. The discovery of the TMPRSS2-ERG fusion sparked an onslaught of discovery and innovation resulting in a delineation of prostate cancer via a molecular signature of gene fusion events. The increased commonality of high-throughput sequencing data coupled with improved bioinformatics approaches not only elucidated the ...
