New Insights into Tumor Suppressor Gene Networks
Special Monday WALS Lecture Cancer arises through an evolutionary process whereby normal cells acquire mutations that erode growth controls, leading to the inappropriate expansion of aberrantly proliferating cells. Such mutations can involve activation of oncogenes or inactivation of tumor-suppressor genes, each contributing one or more new capabilities to the developing cancer cell. Director's Seminar Series Injury and remodeling of blood vessels occur as a direct result of cardiovascular disease, as in atherosclerosis or myocardial infarction, and also in response to therapeutic interventions, including stenting and vein graft bypasses. Dr. Boehm studies vascular injury and remodeling in order to understand the underlying molecular mechanisms and to develop new treatment methods. Mitochondria: dynamic organelles critical for human health
Mitochondria are dynamic organelles that fuse, divide, and move. Human genetic studies indicate that these processes are important for normal functioning of tissues, particularly neurons. Mutations in Mitofusion-2 (Mfn2) cause peripheral neuropathy (Charcot-Marie-Tooth 2A), and mutations in optic atrophy 1 (OPA1) cause eye disease (dominant optic atrophy). Mouse knockout studies focused on the mitofusins Mfn1 and Mfn2 have shown that mitochondrial fusion is important for organellar function. Genomic Networks in Development and Cancer: Resolving Biomarkers and Therapeutic Targets from a Cloud of Data
Systems level approaches to construct abstract molecular networks can lead to predictions about genetic and biochemical functions in cells, organisms and in disease states. I will show examples of this approach from work in my laboratory. In one example we used an integrated experimental and computational approach to construct a large scale functional network in Drosophila melanogaster built around key transcription factors involved in the process of embryonic segmentation.
