Abstract
Within this study, HEC-cysteamine nanoparticles with free thiol groups in the range of 117-1548μmol/g were designed and characterized. Nanoparticles were generated via ionic gelation of the cationic polymer with tripolyphosphate (TPP) followed by covalent crosslinking via disulfide bond formation using H(2)O(2) as oxidant. The mean diameter of the particles was in the range of 270-360nm, and zeta potential was determined to be +4 to +10mV. In this report we describe the development of a novel detection method for the visualization of ligand-binding proteins. Current proteomic tools, such as the Enzyme-linked Immunosorbent Assay (ELISA), are based on protein abundance rather than protein activity and can result in conflicting data. The newly built MagSAXS (magnetic small-angle X-ray scattering) set-up dedicated to the direct two-dimensional measurement of magnetic scattering using polarized synchrotron radiation in extreme sample environments is presented. Pure optical transport of the image is used to record the magnetic scattering with a two-dimensional CCD visible-light camera. The set-up is able to probe magnetic correlation lengths from the micrometer down to the nanometer scale.
