Obesity is rising steadily around the world. Convincing evidence suggests that diet and activity are not the only factors at work in this trend—chemical “obesogens” may alter human metabolism and predispose some people to gain weight. (Source: EHP-in-Press)

Conclusions: Overall, the review of the existing literature identified linkages between several of the environmental exposures and type 2 diabetes. There was also support for the “developmental obesogen” hypothesis, which suggests that chemical exposures may increase the risk of obesity by altering the differentiation of adipocytes or the development of neural circuits that regulate feeding behavior. The effects may be most apparent when the developmental exposure is combined with consumption of a high-calorie, high-carbohydrate, or high-fat diet later in life. Research on environmental chemical exposures and type 1 diabetes was very limited. This lack of research was considered a critical data gap. This workshop report outlines the major themes that emerged from the workshop and discu...

CONCLUSIONS: Adoption studies of ICS or clean energy are scarce, scattered and of differential quality, even while global distribution programs are quickly expanding. Future research must examine an expanded set of contextual variables to improve implementation of stove programs that can realize the “win-win-win” associated with these technologies.
Prioritization of Biomarker Targets in Human Umbilical Cord Blood: Identification of Proteins in Infant Blood Serving as Validated Biomarkers in Adults

Conclusions. This study advanced current knowledge of the human cord blood serum proteome. It showcases the potential of UCB as a diagnostic medium for assessing infant health by detection and identification of candidate biomarkers for known disease pathways using a global, non-targeted approach. These biomarkers may inform on mechanisms of exposure-disease relationships. Furthermore, biomarkers approved by the Food and Drug Administration for screening in adult blood were detected in UCB and represent high-priority targets for immediate validation.
Conclusions: These findings suggest that children who have higher urinary cadmium concentrations may have increased risk of both LD and special education. Importantly, we observed these associations at exposure levels that were previously considered to be without adverse effects and these levels are common among U.S. children.
CONCLUSIONS: Spatial and temporal variability in THM concentrations was greater in water than in blood. We found consistent blood-water correlations across season and site for BDCM and DBCM, and multivariate regression results suggest that water THM concentrations may be an adequate surrogate for baseline blood levels.
Conclusions: Bioaccumulation does not monotonically increase with age. Our model suggests that the main predictors of cross-sectional body burden trends with age are the amount of time elapsed after peak emissions and the human metabolic and environmental degradation rates.
CONCLUSIONS: The global use of DDT has not changed substantially since the Stockholm Convention entered into force. The dominance of pyrethroid use has major implications due to the spread of insecticide resistance with potential to reduce the efficacy of LNs. Insecticide resistance management should be coordinated between disease-specific programs and sectors within the context of an integrated vector management approach.
Conclusions: Prenatal PAH exposure was associated with lower global methylation in umbilical cord white blood cells (p=0.05), but global methylation levels were positively associated with the presence of detectable adducts in cord blood (p=0.01). These observations suggest that PAH exposure was adequate to alter global methylation in our study population. Additional epidemiologic studies that can measure site-specific cytosine methylation and adduct formation will improve our ability to understand this complex molecular pathway in vivo.
Conclusions: The current data demonstrate that PBDE-209 is able to promote proliferation of various cancer cells from the female reproductive system and normal ovarian CHO cells. Furthermore, it reduced tamoxifen, PKCα, and ERK inhibition-induced apoptosis. Finally, PBDE-209 upregulated phosphorylation of PKCα and ERK1/2 proteins in these tumor cells.
Conclusions: The potential adverse effects of in utero PCB exposure on thymic development may extend beyond the neonatal period. Results from this highly exposed cohort provide suggestive evidence that postnatal PCB concentrations may be influential, but a smaller set of 6-month PCB measurements limited statistical power at that time point. Implications regarding impaired immunologic maturation or long-term clinical implications remain to be determined. (Source: EHP-in-Press)

Conclusions. Reducing personal exposure to air pollution using a highly efficient facemask appeared to reduce symptoms and improve a range of cardiovascular health measures in patients with coronary heart disease. Such interventions to reduce personal exposure to particulate air pollution have the potential to reduce the incidence of cardiovascular events in this highly susceptible population.
Editor's Summary

In our daily lives we’re rarely exposed to just one chemical at a time. Metals, for example, are ubiquitous in the environment, and most of us are exposed to different combinations of metals each day through air, water, and food. Simultaneous exposures to different metals may have synergistic effects in children, whose developing brains are particularly vulnerable to adverse effects from these potentially neurotoxic agents. In this podcast host Ashley Ahearn discusses the neurodevelopmental effects of metals mixtures with researcher Robert O. Wright. (Source: EHP-in-Press)

Conclusions: Our findings suggest that exposure to p,p’-DDE may be associated with increased rates of XX, XY, and total sex chromosome disomy, whereas exposure to PCBs may be associated with increased rates of YY, XY, and total sex chromosome disomy. An inverse association was observed between increased exposure to PCBs and XX disomy. Further work is needed to confirm these findings. (Source: EHP-in-Press)

Conclusion: The high levels of environmental contamination, percentage of children aged <5 years with elevated blood-lead levels (97%, >45 µg/dL), and incidence of convulsions among children prior to death (82%) suggest that most of the recent childhood deaths in the two surveyed villages were caused by acute lead poisoning from gold ore-processing activities. Control measures included environmental remediation, chelation therapy, public health education, and control of mining activities. (Source: EHP-in-Press)

Conclusions: We found no significant differences in the serum concentrations of PFOS, PFOA, PFNA, and PFHxS by sex, but increasing concentrations with age. Our results suggest that these 300 Texas children from birth through 12 years of age continued to be exposed to several PFCs in late 2009, years after changes in production of some PFCs in the United States.
Did Smokefree Legislation in England Reduce Exposure to Secondhand Smoke among Non-Smoking adults? Cotinine Analysis from the Health Survey for England

Conclusions: England’s smokefree legislation impacted on secondhand smoke exposure above and beyond the underlying long-term decline demonstrating the positive effect of the legislation. Nevertheless, some population subgroups appear not to have benefitted significantly from the legislation, suggesting that these groups should receive more support to reduce their exposure.
Conclusions: These results suggest that Pb exposure during childhood impairs the child’s ability to allocate the cognitive resources needed to correctly inhibit a prepotent response, resulting in increased impulsivity. By contrast, postnatal PCB exposure appears to affect processes associated with error monitoring, an aspect of behavioural regulation required to adequately adapt to the changing demands of the environment, which results in reduced task efficiency.
Conclusions: O3 exposure causes several alterations in cardiac electrophysiology that are likely mediated by modulation of autonomic input to the heart. Moreover, exposure to low O3 concentrations may cause subclinical effects that are only manifested when triggered by a stressor, suggesting that the health effects of ambient levels of air pollutants may be insidious and potentially underestimated (This abstract does not reflect EPA policy).
Conclusion: This systematic review identified an association between arsenic and the prevalence of hypertension. Interpreting a causal effect of arsenic on hypertension is limited by the small number of studies, the presence of influential studies and the absence of prospective evidence. Additional evidence is needed to evaluate the dose-response relationship between arsenic exposure and hypertension. (Source: EHP-in-Press)

Editor's Summary

With more than 1 billion people estimated to not have enough to eat, food security is a pervasive problem. An estimated one-third of the global burden of disease afflicting children under the age of 5 is caused by undernutrition. Climate change is anticipated to reduce cereal yields, further threatening food security and potentially increasing child undernutrition. In this podcast, host Ashley Ahearn discusses the connection between climate change and undernutrition with researcher Sari Kovats. (Source: EHP-in-Press)

CONCLUSIONS: Public health nurses view the environment as under threat and see a role for nursing divisions in addressing health effects of climate change. However, they recognize the limited resources and personnel available to devote to this endeavor. (Source: EHP-in-Press)

Conclusions: Expert responses were generally consistent with the peer-reviewed literature regarding the relationship between climate change and vector- and waterborne diseases, but were less so for foodborne diseases. Shortcomings in institutional capacity to manage climate change vulnerability, identified in this assessment, should be addressed with impact, vulnerability and adaptation assessments.
Conclusions: Occupational exposures to benzene were associated with increased incidence of chromosomally defective sperm raising concerns for worker infertility, spontaneous abortions, as well as mental retardation and inherited defects in their children. Our sperm findings point to benzene as a possible risk factor for de novo 1p36 deletion syndrome. Because chromosomal aberrations in sperm can arise from defective stem cells/spermatogonia, our findings raise concerns that occupational exposure to benzene may have persistent reproductive effects in formerly exposed workers.
Reduction in Cadmium Exposure in the United States Population, 1988-2008: The Contribution of Declining Smoking Rates

Conclusions. Urine cadmium concentrations decreased markedly between 1988 and 2008. Declining smoking rates and changes in exposure to tobacco smoke may have played an important role in the decline of urine cadmium concentrations, benefitting both smokers and non-smokers. Cadmium has been associated to several health outcomes in NHANES 1999-2008. Consequently, despite the observed decline, further reduction in cadmium exposure is needed.
Conclusions: The high prevalence (61%) of lead above recommended levels in soil and dust samples in and around residences raises concern about potential health risks to the New Orleans population, most notably children. Steps should be taken to mitigate the risk of exposure to lead-contaminated soil and dust. Further research is needed to quantify the possible contribution of reconstruction activities to environmental lead levels. (Source: EHP-in-Press)

Conclusions: With the exception of monozygotic multiple birth and maternal hypertensive disorder, early-life risk factors for early onset fibroids in blacks were similar to those in whites. However in contrast to whites, childhood height and weight, but not low socioeconomic status indicators, were associated with early onset fibroids in blacks. The general consistency of early-life findings for blacks and whites supports a possible role of early-life factors in fibroid development. (Source: EHP-in-Press)

Lesson Summary

How do genotoxins damage DNA, and how does the cell repair the damage? Students create models to predict how genotoxicants can cause DNA damage and learn about the different ways in which a cell repairs this damage. Students also summarize the abstract of an EHP research article Transcription and DNA Damage: A Link to A Kink, and learn how DNA damage repair plays an important role in DNA transcription.
Lesson Summary

Why do physical characteristics of identical twins diverge with age? Students learn that our environment can alter the way our genes are expressed, making even identical twins different. After watching a PBS video A Tale of Two Mice, and reviewing data presented in the EHP article Maternal Genistein Alters Coat Color and Protects Avy Mouse Offspring from Obesity by Modifying the Fetal Epigenome, students learn about epigenetics and its role in regulating gene expression.
CONCLUSIONS: Variations in gut microbiota are likely to affect human toxicodynamics and increase individual exposure to obesogenic and diabetogenic chemicals. Combating the global obesity/diabetes epidemics requires a multifaceted approach that should include greater emphasis on understanding and controlling the impact of interindividual gut microbe variability on the disposition of environmental chemicals in humans.
Conclusions: Results suggest that a Cd-mediated increase in urinary albumin excretion is reversible upon substantial reduction of exposure. For the markers of tubular effects, a tendency towards improvement, but not complete recovery, was observed. Data from repeated observations suggests that β2-microglobulin may be more informative than NAG as an indicator for the individual’s future tubular function. (Source: EHP-in-Press)

Background: Physical inactivity and exposure to air pollution are important risk factors for death and disease globally. The built environment may influence exposures to these risk factors in different ways and thus differentially impact the health of urban populations.
Objective: To investigate the built environment’s association with air pollution and physical inactivity, and estimate attributable health risks.
Methods: We used a regional travel survey to estimate within-urban variability in physical inactivity and home-based air pollution exposure (PM2.5, NOx, O3) for 30,007 individuals in southern California. We then estimated the resulting risk for ischemic heart disease (IHD) using literature-derived dose-response values for the general public and for a sensitive subpopulation. Usi...

Conclusion: These data show that 1,2-NQ exposure induces mitochondrial production of H2O2
that mediates the expression of inflammatory genes, but not the concurrent loss of reducing
redox potential in BEAS cells. 1,2-NQ exposure also causes marked expression of HO-1 that
appears to be enhanced by suppression of H2O2. These findings shed light into the oxidantdependent events that underlie cellular responses to environmental electrophiles. (Source: EHP-in-Press)

This study investigated the role of chronic, oral exposure to BPA during adulthood on mammary carcinogenesis by using a transgenic mouse model which spontaneously develops tumors through over-expression of wildtype erbB2 (MMTV-erbB2).
Methods: MMTV-erbB2 mice were exposed to 0, 2.5, 25, 250, or 2500 µg BPA/L drinking water from 56 until 112 (mechanism of action) or 252 (tumorigenesis) days of age. Cellular and molecular mechanisms of BPA action in the mammary gland were investigated via immunohistochemistry and immunoblotting.
Results: Only low doses of BPA significantly decreased tumor latency and increased tumor multiplicity, tumor burden, and the incidence of metastasis. All BPA doses significantly increased the cell proliferation index, but only the higher doses also increased the apo...
