Lessons from a systematic literature search on diagnostic DNA methylation biomarkers for colorectal cancer: how to increase research value and decrease research waste
DiscussionThis systematic literature search revealed that major requirements to develop clinically relevant diagnostic CRC DNA methylation markers are often lacking. To avoid the resulting research waste, clinical needs, intended biomarker use and independent validation should be better considered prior to study design. In addition, improved reporting quality would facilitate meta-analysis, thereby increasing level of evidence and enabling clinical translation.PMID:35584320 | DOI:10.14309/ctg.0000000000000499

Relationship Between State-Level Google Online Search Volume and Cancer Incidence in the United States: Retrospective Study
Conclusions: Cancer incidence is correlated with online search volume at the state level. Search patterns were temporally associated with cancer awareness months and celebrity announcements. Online searches reflect public awareness. Advancing understanding of online search patterns could augment traditional epidemiologic surveillance, provide opportunities for targeted patient engagement, and allow public information campaigns to be evaluated in ways previously unable to be measured.

Interventions for weight reduction in obesity to improve survival in women with endometrial cancer
CONCLUSIONS: The inclusion of new relevant studies has not changed the conclusions of this review. There is currently insufficient high-quality evidence to determine the effect of combined lifestyle and behavioural interventions on survival, quality of life or significant weight loss in women with a history of endometrial cancer who are overweight or obese compared to those receiving usual care. The limited evidence suggests that there is little or no serious or life-threatening adverse effects due to these interventions, and it is uncertain if musculoskeletal problems were increased, as only one out of eight studies repor...

Hyperbaric oxygen therapy for late radiation tissue injury
CONCLUSIONS: These small studies suggest that for people with LRTI affecting tissues of the head, neck, bladder and rectum, HBOT may be associated with improved outcomes (low- to moderate-certainty evidence). HBOT may also result in a reduced risk of wound dehiscence and a modest reduction in pain following head and neck irradiation. However, HBOT is unlikely to influence the risk of death in the short term. HBOT also carries a risk of adverse events, including an increased risk of a reduction in visual acuity (usually temporary) and of ear barotrauma on compression. Hence, the application of HBOT to selected participants ...

Screening for prostate cancer.
CONCLUSIONS: Prostate cancer screening did not significantly decrease prostate cancer-specific mortality in a combined meta-analysis of five RCTs. Only one study (ERSPC) reported a 21% significant reduction of prostate cancer-specific mortality in a pre-specified subgroup of men aged 55 to 69 years. Pooled data currently demonstrates no significant reduction in prostate cancer-specific and overall mortality. Harms associated with PSA-based screening and subsequent diagnostic evaluations are frequent, and moderate in severity. Overdiagnosis and overtreatment are common and are associated with treatment-related harms. Men sh...

The Prognostic and Clinical Value of CD44 in Colorectal Cancer: A Meta-Analysis
Conclusions: This meta-analysis demonstrated that CD44 overexpression might be an unfavorable prognostic factor for CRC patients and could be used to predict poor differentiation, lymph node metastasis and distant metastasis. Introduction Although therapies for colorectal cancer (CRC) has improved in recent years, colorectal cancer is still the third most common cause of cancer related death worldwide (1). Metastasis are observed in 25% of patients at initial diagnosis and approximately 50% of patients will develop metastasis (2). Presently, the outcome prediction and the therapy schedule determination of CRC patie...

Systematic Search for Present and Potential Portals of Entry for Infective Endocarditis
ConclusionsOur search for the POEs of present IEs was often successful, as was searching for an oral or dental or a gastrointestinal POE of a new IE episode. We advise the systematic performance of stomatologic examinations in patients with IE and performance of colonoscopy in patients ≥50 years of age or at high risk for colorectal cancer.

Abstract 5555: Pharmacogenetics-pharmacokinetics study of bevacizumab in mCRC patients treated with Avastin-Folfiri regimen: Search for predictive markers of response and study of the pharmacokinetics variability
Identifying biomarkers predictive of response is a rising trend in clinical oncology, especially when administrating costly targeted therapies and biologics. Unlike other monoclonal antibodies such as cetuximab, panitumumab, trastuzumab or ipilimumab, search for genetic or molecular makers predictive for clinical outcome with widely prescribed bevacizumab has failed to generate consensual results over the last decade. We have performed a pilot study in 70 patients with metastatic colorectal cancer receiving the standard bevacizumab-Folfiri protocol. More specifically, our study focused on the retrospective screening of the...

Abstract CT211: Biomarker search using gene expression databases in a phase III controlled clinical trial of postoperative adjuvant chemotherapy for stage III colon cancer (ACTS-CC): correlation between clinicopathological factors and gene expression level
Conclusions: Location, differentiation and depth of the tumor correlated with each gene expressions. Our findings will facilitate understanding the characteristics of colon cancer. Further investigation on gene expressions including genome-wide copy number analysis may contribute to the exploration of valid biomarkers.Citation Format: Hiroyuki Uetake, Toshiaki Ishikawa, Megimi Ishiguro, Shigeyuki Matsui. Role of "Second Look" Lymph Node Search in Harvesting Optimal Number of Lymph Nodes for Staging of Colorectal Carcinoma.
Authors: Hamza A, Sakhi R, Khawar S, Alrajjal A, Edens J, Khurram MS, Khan U, Szpunar S, Mazzara P Abstract As with other malignancies, lymph node metastasis is an important staging element and prognostic factor in colorectal carcinomas. The number of involved lymph nodes is directly related to decreased 5-year overall survival for all pT stages according to United States Surveillance, Epidemiology, and End Results (SEER) cancer registry database. The National Quality Forum specifies that the presence of at least 12 lymph nodes in a surgical resection is one of the key quality measures for the evaluation of colorec...

Cancers, Vol. 12, Pages 3311: Role of miRNAs in Sigmoid Colon Cancer: A Search for Potential Biomarkers
a Ferreira Vidal Vivian Nogueira Silbiger Ândrea Ribeiro-dos-Santos The aberrant expression of microRNAs in known to play a crucial role in carcinogenesis. Here, we evaluated the miRNA expression profile of sigmoid colon cancer (SCC) compared to adjacent-to-tumor (ADJ) and sigmoid colon healthy (SCH) tissues obtained from colon biopsy extracted from Brazilian patients. Comparisons were performed between each group separately, considering as significant p-values < 0.05 and |Log2(Fold-Change)| > 2. We found 20 differentially expressed miRNAs (DEmiRNAs) in all comparisons, two of which were shar...
