Simvastatin, but Not Atorvastatin, Is Associated with Higher Peak Rivaroxaban Serum Levels and Bleeding: an Asian Cohort Study from Singapore
ConclusionOur findings suggest that concomitant use of simvastatin with rivaroxaban may be associated with bleeding events in an Asian cohort. Further studies using physiologically based pharmacokinetic modelling are required to investigate the drug-drug interactions between these drugs.

Influence of non-osteoporotic treatments in patients on active anti-osteoporotic therapy: evidence from the OSTEOMED registry
ConclusionThe potential beneficial or detrimental effects of the different drugs evaluated on fracture risk are masked by treatment with anabolic or antiresorptive drugs that have a more potent action on bone metabolism, with two exceptions: letrozole and oral corticosteroids. These findings may have important clinical implications, as patients receiving these treatments are not fully protected by bisphosphonates, which may imply the need for more potent anti-osteoporotic drugs such as denosumab or teriparatide.

The effects of statins on the function and differentiation of blood cells
Arch Med Sci. 2022 Dec 25;19(5):1314-1326. doi: 10.5114/aoms/158546. eCollection 2023.ABSTRACTStatins are inhibitors of β-hydroxy β-methylglutaryl-CoA (HMG-CoA) reductase (HMGCR). They are used in patients with cardiovascular risk and/or suffering with cardiovascular disease. In addition to their efficient lipid-lowering effects, statins exhibit independent so called pleiotropic effects potentially affecting several immune response properties including immune cell activation, migration, cytokine generation, immune metabolism, and survival. Statins also regulate innate and acquired immunity. The focus of this review is to...

Preventive effects of 13 different drugs on colorectal cancer: a network meta-analysis
CONCLUSIONS: The preventive effect of allopurinol on CRC was better than that of the other 13 drugs. These results can help doctors and patients understand the preventative effects of these drugs more intuitively and provide an evidence-based basis for the clinical application of these drugs.PMID:37732038 | PMC:PMC10507769 | DOI:10.5114/aoms/167480

A cardiovascular polypill for secondary stroke prevention in a tertiary centre in Ghana (SMAART): a phase 2 randomised clinical trial
Lancet Glob Health. 2023 Oct;11(10):e1619-e1628. doi: 10.1016/S2214-109X(23)00347-9.ABSTRACTBACKGROUND: A cardiovascular polypill containing generic drugs might facilitate sustained implementation of and adherence to evidence-based treatments, especially in resource-limited settings. However, the impact of a cardiovascular polypill in mitigating atherosclerotic risk among stroke survivors has not been assessed. We aimed to compare a polypill regimen with usual care on carotid intima-media thickness (CIMT) regression after ischaemic stroke.METHODS: In SMAART, a phase 2 parallel, open-label, assessor-masked, randomised clini...

Quantifying the impact of taking medicines for primary prevention: a time-trade off study to elicit direct treatment disutility in the UK
Conclusions Statins and bisphosphonates have a quantifiable DTD. The size of estimated disutilities suggest they are likely to be important for cost-effectiveness, particularly in individuals at low-risk when treated for primary prevention.

Frequency of lipoprotein(a) measurements in patients with or at risk of cardiovascular disease
Lipoprotein (a) [Lp(a)] is an apolipoprotein B-containing low density lipoprotein with an additional apolipoprotein(a) bound. It is a highly prevalent, genetically determined, biomarker strongly predicting atherosclerotic cardiovascular disease (ASCVD) events and calcific aortic stenosis.1 Lp(a) contributes to cardiovascular disease through pro-atherogenic, pro-inflammatory, and pro-thrombotic effects.2,3 A meta-analysis of statin-treated patients with an elevated baseline and on-statin lipoprotein(a) showed a linear relationship with cardiovascular risk.
