Pharmacological treatment options for fibromyalgia
Fibromyalgia is a high-impact and prevalent condition. Its clinical features include widespread pain and tenderness — often associated with high levels of poor-quality sleep, fatigue and cognitive dysfunction. The mechanism leading to fibromyalgia development is associated with a change in the sensitivity of the pain-related nervous system, as well as other sensory systems. In turn, emotional distress plays a facilitatory role in this process. Non-pharmacological management focuses ...

Nerve growth factor inhibitor with novel ‐binding domain demonstrates nanomolar efficacy in both cell‐based and cell‐free assay systems
Abstract Nerve growth factor (NGF), a member of the neurotrophin family, is known to regulate the development and survival of a select population of neurons through the binding and activation of the TrkA receptor. Elevated levels of NGF have been associated with painful pathologies such as diabetic neuropathy and fibromyalgia. However, completely inhibiting the NGF signal could hold significant side effects, such as those observed in a genetic condition called congenital insensitivity to pain and anhidrosis (CIPA). Previous methods of screening for NGF‐inhibitors used labeling techniques which have the potential to alter...

Early Use of Ceftaroline Fosamil in the United States Veterans Health Care System
ConclusionsCeftaroline is used in VHA hospitals for various diagnoses. Mortality was low and comparable with rates from clinical trials. Additional studies comparing ceftaroline to other drugs used in similar situations are needed.

Efficacy of Venlafaxine in Neuropathic Pain: A Narrative Review of Optimized Treatment.
We describe the 13 studies that we analyzed. FINDINGS: Eleven were randomized clinical trials, and the comparator was placebo in 8 studies. Nine studies reported that VLX was effective against neuropathic pain. However, among the trials, only one against placebo included a large number of patients with >200 participants and one against prégabaline and carbamazepine had >200 patients. Most of the adverse events reported in the selected studies were consistent with known adverse events of VLX, and most were mild to moderate. However, most studies were of very short duration. IMPLICATIONS: Most of the clinica...

Anti-hyperalgesic effect of Lippia grata leaf essential oil complexed with β-cyclodextrin in a chronic musculoskeletal pain animal model: Complemented with a molecular docking and antioxidant screening.
CONCLUSION: This study suggested that LG-βCD could be considered as a valuable source for designing new drugs in the treatment of chronic pain, especially musculoskeletal pain. PMID: 28499245 [PubMed - as supplied by publisher]

From acute to chronic pain: tapentadol in the progressive stages of this disease entity.
CONCLUSIONS: Tapentadol represents a suitable candidate for patients at early progressive stage of pain who have developed neuroplasticity with modification of pain pathways. The availability of different doses of tapentadol may help clinicians to tailor treatment based on the individual need of each patient, with the aim to enhance therapeutic appropriateness in the treatment of musculoskeletal and neuropathic pain. Low ‐dose naltrexone and opioid consumption: a drug utilization cohort study based on data from the Norwegian prescription database
ConclusionsPossibly, LDN users avoided opioids because of warnings on concomitant use or the patients continuing on LDN were less opioid dependent than those terminating LDN. Therapeutic effects of LDN contributing to lower opioid consumption cannot be ruled out. © 2017 The Authors. Pharmacoepidemiology & Drug Safety Published by John Wiley & Sons Ltd.

The role of oxidative stress in the relation between fibromyalgia and obstructive sleep apnea syndrome.
CONCLUSIONS: OSAS and FMS were highly prevalent, which indicated that oxidative stress might play a role in the pathophysiology of both diseases, especially if they co-exist in the same patient. PMID: 28121361 [PubMed - in process]

Repeated Cold Stress Enhances the Acute Restraint Stress-Induced Hyperthermia in Mice.
Abstract The rodents exposed to repeated cold stress according to a specific schedule, known as specific alternation of rhythm in temperature (SART), exhibit autonomic imbalance, and is now used as an experimental model of fibromyalgia. To explore the susceptibility of SART-stressed animals to novel acute stress, we tested whether exposure of mice to SART stress for 1 week alters the extent of acute restraint stress-induced hyperthermia. Mice were subjected to 7-d SART stress sessions; i.e., the mice were alternately exposed to 24 and 4°C at 1-h intervals during the daytime (09:00-16:00) and kept at 4°C overnigh...

Enhanced Hyperthermic Responses to Lipopolysaccharide in Mice Exposed to Repeated Cold Stress
Lipopolysaccharide (LPS) induces hyperthermia accompanied by various other systemic inflammatory symptoms. The rodents exposed to repeated cold (RC) stress according to a specific schedule are useful as experimental models for autonomic imbalance or fibromyalgia. It is now proven that RC-stressed mice exhibit tolerance to LPS, we examined thermal responses to LPS challenge in RC-stressed mice by monitoring core temperature using the telemetry system. Systemic administration of LPS caused bimodal hyperthermic responses in RC-stressed and unstressed mice. The magnitude of the LPS-induced hyperthermia was greater in RC-stress...

The safety of pregabalin in the treatment of fibromyalgia.
This article reviews pregabalin's safety profile. Areas covered: The safety findings in pregabalin clinical trials were accessed by a PubMed search using the key words 'pregabalin' or 'anti-epilectics drug' or 'gabapentinoids' or 'anticonvulsants' and 'fibromyalgia'. Although frequent, the side effects of pregabalin therapy are usually mild to moderate, well tolerated in the long term, and can be monitored in a primary care setting. Expert opinion: Pregabalin therapy may be associated with somnolence, dizziness, weight gain and periphereal edema. Potential drug interactions are not common, and pregabalin seems to be well t...
