Off-target effects of oral anticoagulants - vascular effects of vitamin K antagonist and non-vitamin K antagonist oral anticoagulant dabigatran etexilate
CONCLUSION: Warfarin aggravates atherosclerotic disease activity, increasing plaque inflammation, active calcification and plaque progression. Dabigatran lacks undesired vascular side effects and reveals beneficial effects on atherosclerosis progression and calcification. The choice of anticoagulation impacts atherosclerotic disease by differential off target effect. Future clinical studies should test whether this beneficial effect also applies to patients.PMID:33687782 | DOI:10.1111/jth.15289

Review: Circadian clocks and rhythms in the vascular tree
Curr Opin Pharmacol. 2021 Jun 7;59:52-60. doi: 10.1016/j.coph.2021.04.010. Online ahead of print.ABSTRACTThe progression of vascular disease is influenced by many factors including aging, gender, diet, hypertension, and poor sleep. The intrinsic vascular circadian clock and the timing it imparts on the vasculature both conditions and is conditioned by all these variables. Circadian rhythms and their molecular components are rhythmically cycling in each endothelial cell, smooth muscle cell, in each artery, arteriole, vein, venule, and capillary. New research continues to tackle how circadian clocks act in the vasculature, d...

Small molecule compound M12 reduces vascular permeability in obese mice via blocking endothelial TRPV4-Nox2 interaction
In this study we investigated the function of TRPV4-Nox2 complex in reducing oxidative stress and regulating abnormal vascular permeability in obesity. Obesity was induced in mice by feeding a high-fat diet (HFD) for 14 weeks. The physical interaction between TRPV4 and Nox2 was measured using FRET, co-immunoprecipitation and GST pull-down assays. The functional interaction was measured by rhodamine phalloidin, CM-H2DCFDA in vitro, the fluorescent dye dihydroethidium (DHE) staining assay, and the Evans blue permeability assay in vivo. We demonstrated that TRPV4 physically and functionally associated with Nox2, and this phys...

The epigenetic enzyme DOT1L orchestrates vascular smooth muscle cell-monocyte crosstalk and protects against atherosclerosis via the NF-kappaB pathway
CONCLUSION: DOT1L plays a key role in the epigenetic control of VSMC gene expression, leading to atherosclerosis development. Results identify DOT1L as a potential therapeutic target for vascular diseases.KEY QUESTION: Epigenetics plays an important role in the regulation of atherosclerosis development. Its role in vascular smooth muscle cell biology and immune cell recruitment is not yet fully understood.KEY FINDING: Inhibition of the epigenetic enzyme disruptor of telomeric silencing 1-like (DOT1L) in vascular smooth muscle cells significantly reduces atherosclerosis progression, directly modulating Nfκb1 and Nfκb2 tra...

Effects of dietary palmitoleic acid on vascular function in aorta of diabetic mice
CONCLUSION: These results suggest that POA intake enhances vasoconstriction under hyperglycemic and hyperinsulinemic conditions, which are characteristics of type 2 diabetes, and may contribute to increased vascular complications in diabetes.PMID:35436932 | DOI:10.1186/s12902-022-01018-2
