Wilms tumor: Kidney cancer in children
While kidney cancer is rare in children, Wilms tumor is the most common type found in kids, with between 500 and 600 children diagnosed annually in the U.S. The disease most often affects kids ages 3 to 4. Thanks to advances in treatments over the past several decades, survival rates from this rare cancer have improved significantly. Developing a treatment plan customized for each child with Wilms tumor has also helped to improve the prognosis for children… (Source: News from Mayo Clinic)

Consumer Health: What do you know about these 3 rare childhood cancers?
Childhood cancer rates have been rising slightly for the past few decades, and about 10,470 children in the U.S. under the age of 15 will be diagnosed with cancer in 2022, according to the American Cancer Society. September is Childhood Cancer Awareness Month, which makes this a good time to learn about three rare types of cancers in children: retinoblastoma, rhabdomyosarcoma and Wilms' tumor. Retinoblastoma Retinoblastoma is a rare form of eye cancer that begins in the… (Source: News from Mayo Clinic)

New NCCN guidelines analyze evidence for cancer type found almost exclusively in children
(National Comprehensive Cancer Network) National Comprehensive Cancer Network publishes first-ever complete medical guidelines for a pediatric solid tumor, so children everywhere receive the best care based on the latest research. NCCN Guidelines for Wilms Tumor shares information for earlier, safer diagnosis and treatment in effort to reduce disparities; doctors warn of late diagnosis as a possible impact from the COVID-19 pandemic. Root of childhood kidney cancer discovered
(Wellcome Trust Sanger Institute) A fundamental change in our understanding of the childhood kidney cancer Wilms' tumor is on the horizon, after the discovery of its earliest genetic root by scientists at the Wellcome Sanger Institute and their collaborators. By comparing genome sequences from normal kidney tissue and tumors, the team identified patches of normal-looking kidney tissue that in fact carried DNA changes that cause Wilms' tumor. Embryonal precursors of Wilms tumor
Adult cancers often arise from premalignant clonal expansions. Whether the same is true of childhood tumors has been unclear. To investigate whether Wilms tumor (nephroblastoma; a childhood kidney cancer) develops from a premalignant background, we examined the phylogenetic relationship between tumors and corresponding normal tissues. In 14 of 23 cases studied (61%), we found premalignant clonal expansions in morphologically normal kidney tissues that preceded tumor development. These clonal expansions were defined by somatic mutations shared between tumor and normal tissues but absent from blood cells. We also found hyper...

Single-cell transcriptomes from human kidneys reveal the cellular identity of renal tumors
Messenger RNA encodes cellular function and phenotype. In the context of human cancer, it defines the identities of malignant cells and the diversity of tumor tissue. We studied 72,501 single-cell transcriptomes of human renal tumors and normal tissue from fetal, pediatric, and adult kidneys. We matched childhood Wilms tumor with specific fetal cell types, thus providing evidence for the hypothesis that Wilms tumor cells are aberrant fetal cells. In adult renal cell carcinoma, we identified a canonical cancer transcriptome that matched a little-known subtype of proximal convoluted tubular cell. Analyses of the tumor compos...

Scientists Identify New Mechanisms Underlying Pediatric Kidney Cancer
In a new study, researchers have found that miRNA processing mutations exhibit higher levels of Pleomorphic adenoma gene 1 (PLAG1), a protein that normally functions during development to turn on IGF2 expression. In kidney cells with miRNA mutations, PLAG1 levels are abnormally high, leading to inappropriate production of IGF2, which in turn promotes Wilms tumor formation.08/02/2018 (Source: Kidney Cancer Association)

Should I Worry About This Telangiectasia?
Discussion Vascular stains are common problems that parents seek guidance about as they are often particularly worried that there may be an underlying problem or that it may be a long-term cosmetic problem. Fortunately many resolve or become less prominent or have treatment available. The term vascular stains includes all vascular malformations but commonly refers more directly to capillary malformations which are quite common. Some common vascular stains include: Nevus simplex Names: Angel’s kiss (glabella or forehead), salmon patch, stork bite (nape of neck), nevus roseus, fading macular stain Epidemiology: Very ...
