Non-detection of Epstein-Barr virus and human papillomavirus in a region of high gastric cancer risk indicates a lack of a role for these viruses in gastric carcinomas
Gastric mucosa tissue was collected from patients with gastroduodenal diseases in a region of norrteastern China showing a high risk of gastric cancer incidence. The presence of EBV and HPV were assayed to investigate the relationship between gastric carcinomas and virus infection. Neither EBV nor HPV DNA was detected in tissue from the patients. The role of EBV and HPV in gastric cancer is not well understood and still needs to be clarified. (Source: Genetics and Molecular Biology)

MiR-204 down regulates SIRT1 and reverts SIRT1-induced epithelial-mesenchymal transition, anoikis resistance and invasion in gastric cancer cells
Conclusion: Our results suggest that down-regulation of miR-204 promotes gastric cancer cell invasion by activating the SIRT1-LKB1 pathway. These data demonstrate that miR-204 plays an important role in regulating metastasis of gastric cancer, which is involved in post-transcriptional repression of SIRT1. (Source: BMC Cancer)

Protein expression status in mucosal and submucosal portions of early gastric cancers and their predictive value for lymph node metastasis
We aimed to find out predictive markers for lymph node (LN) metastasis of early gastric carcinoma (EGC) by separating evaluation of protein expression in mucosa and submucosa considering tumor heterogeneity. We selected 37 pN1–3 EGCs and depth‐ and size‐matched 31 pN0 EGCs as training set and 72 EGCs including 14 pN1–3 EGCs as test set. Protein expression for β‐catenin, E‐cadherin, N‐cadherin, galectin‐3, c‐MET, TrkB, and Ki‐67 was assessed by immunohistochemistry in mucosal (‐m) and submucosal (‐sm) portions of tumor. In the training set, Ki67‐m was higher than in Ki67‐sm (mean ± SD: 82.67...

CDH1 germline mutations and the hereditary diffuse gastric and lobular breast cancer syndrome: a multicentre study
We report all index cases from Ile-de-France in which a germline CDH1 mutation has been identified. Out of 18 cases, 7 do not fulfil the HDGC-defining criteria. Three of them are women who presented initially with bilateral LBC below age 50, without personal or family history of DGC, and who subsequently developed symptomatic DGC. Discussion Our series of CDH1 mutation carriers is the largest to date and demonstrates that LBC might be the first manifestation of HDGC. A personal or family history of multiple LBCs at a young age, even without DGC, should prompt CDH1 mutation screening. It is paramount to identify mutation c...

Systematic Review of Studies Investigating Sentinel Node Navigation Surgery and Lymphatic Mapping for Gastric Cancer.
Abstract Abstract Evolutions in minimally invasive surgical techniques and advances in sentinel node navigation surgery (SNNS) have had considerable impact on current insights into surgical treatment of gastric cancer. Extensive data on this method of surgical application have accumulated but have been principally derived from single-institute studies. Isosulfan blue and patent blue violet have been the most frequently used dye tracers in the past; however, indocyanine green has now is increasingly popular. The double-tracer method, where dye and radioisotope tracers are used together, seems to be more effective th...

Diabetes drugs may be linked to pancreatic cancer
Conclusion This article presents important concerns that glucagon-like peptide-1 (GLP-1) agonists and dipeptidylpeptidase-4 (DPP-4) inhibitors could potentially increase the risk of inflammation and cancerous changes in the pancreas. The agencies that regulate medicines in Europe and the USA are aware of these issues, and told the BMJ that their analyses show increased reporting of pancreatic cancer among people taking these types of drugs. However, the agencies note that it has not been established whether these drugs directly cause the adverse effects seen in the pancreas. Both agencies are reviewing emerging eviden...

Phase II/III multicentre randomised controlled trial evaluating a strategy of primary surgery and adjuvant chemotherapy versus peri-operative chemotherapy for resectable gastric signet ring cell adenocarcinomas -- PRODIGE 19 -- FFCD1103 -- ADCI002
DiscussionAs inherent chemo resistance of signet ring cell tumours and delay in definitive surgery may favour tumour progression we hypothesise that a policy of primary surgery followed by adjuvant chemotherapy will improve overall survival compared to a standard perioperative chemotherapeutic strategy. This randomised phase II/III trial is the first dedicated to this histological subtype. Whilst the development of new biomarkers and targeted therapies are awaited, the results of this trial should further help in devising individualised protocols of patient care in a tumour group whose diversity increasingly demands assess...

E2F transcription factors and digestive system malignancies: How much do we know?
Abstract E2F family of transcription factors regulates various cellular functions related to cell cycle and apoptosis. Its individual members have traditionally been classified into activators and repressors, based on in vitro studies. However their contribution in human cancer is more complicated and difficult to predict. We review current knowledge on the expression of E2Fs in digestive system malignancies and its clinical implications for patient prognosis and treatment. E2F1, the most extensively studied member and the only one with prognostic value, exhibits a tumor-suppressing activity in esophageal, gastric ...

Short-course radiotherapy followed by neo-adjuvant chemotherapy in locally advanced rectal cancer ¿ the RAPIDO trial
DiscussionFollowing the advances in rectal cancer management, increased focus on survival rather than only on local control is now justified. In an experimental arm, short-course radiotherapy is combined with full-dose chemotherapy preoperatively, an alternative that offers advantages compared to concomitant chemoradiotherapy with or without postoperative chemotherapy. In a multi-centre setting this regimen is compared to current standard with the aim of improving survival for patients with locally advanced rectal cancer.Trial registrationClinicalTrials.gov NCT01558921 (Source: BMC Cancer)

Red-Flag Technologies in Gastric Neoplasia
This article reviews the emerging use of enhanced endoscopic imaging technologies in the detection and management of gastric cancer. The combined use of white-light endoscopy with enhanced imaging technologies, such as magnification narrow-band imaging, chromoendoscopy, and autofluorescence endoscopy, demonstrates promise in the improved ability to detect and delineate gastric neoplasia. However, widespread clinical use is still limited, mainly because of the restricted availability of the technologies. (Source: Gastrointestinal Endoscopy Clinics of North America)

Sporadic fundic gland polyps and proximal polyposis associated with gastric adenocarcinoma share a common antral G cell hyperplasia
We read with great interest the article by Worthley et al1 that recently appeared in your Journal, and the subsequent letter on the same subject.2 We would like to make some comments on this interesting topic. Worthley et al reported a new autosomal dominant syndrome that they named gastric adenocarcinoma and proximal polyposis of the stomach (GAPPS). GAPPS appears to be characterised by proximal gastric polyposis, particularly of the fundic gland type that often displays low and high grade dysplasia with early development of gastric cancer. These patients showed no evidence of colonic polyps, and other genetic syndromes w...

Time trends of cancer incidence and mortality in Catalonia during 1993-2007.
CONCLUSION: Among men, the decrease of incidence/mortality of tobacco-related tumors was related to a reduction of smoking prevalence. Among women, the stabilization of breast cancer incidence and the rise of lung cancer incidence are similar to that observed in most European regions. The Helicobacter pylori Protein CagM is Located in the Transmembrane Channel That is Required for CagA Translocation.
Abstract Helicobacter pylori (H. pylori) is a human gastric pathogen that colonizes the stomach in more than 50 % of the world's human population. Infection with this bacterium can induce several gastric diseases ranging from gastritis to peptic ulcer and gastric cancer. Virulent H. pylori isolates harboring the cag pathogenicity island (cag PAI), which encodes a Type IV Secretion System (T4SS), form a pilus for the injection of its major virulence protein CagA into gastric cells. Several cag PAI genes have been identified as homologues of T4SS genes from Agrobacterium tumefaciens, while the other members in cag P...

Gastric carcinoma: imaging diagnosis, staging and assessment of treatment response.
Abstract Gastric carcinoma (GC) is one of the most common causes of cancer-related death worldwide. Surgical resection is the only cure available and is dependent on the GC stage at presentation, which incorporates depth of tumor invasion, extent of lymph node and distant metastases. Accurate preoperative staging is therefore essential for optimal surgical management with consideration of preoperative and/or postoperative chemotherapy. Multidetector computed tomography (MDCT) with its ability to assess tumor depth, nodal disease and metastases is the preferred technique for staging GC. Endoscopic ultrasonography is...

Silencing of Ghrelin Receptor Expression Inhibits Endometrial Cancer Cell Growth in vitro and in vivo.
Abstract Ghrelin is a 28 amino acid peptide hormone produced predominantly in the stomach, but also in a range of normal cell types and tumours, where it has endocrine, paracrine, and autocrine roles. We have previously demonstrated that ghrelin has proliferative and anti-apoptotic effects in endometrial cancer cell lines, suggesting a potential role in promoting tumour growth. In the present study, we investigated the effect of ghrelin receptor, GHSR, gene silencing in vitro and in vivo, and characterized ghrelin and GHSR1a protein expression in human endometrial tumours. GHSR gene silencing was achieved in the Is...

Comprehensive genome- and transcriptome-wide analyses of mutations associated with microsatellite instability in Korean gastric cancers [RESEARCH]
Microsatellite instability (MSI) is a critical mechanism that drives genetic aberrations in cancer. To identify the entire MS mutation, we performed the first comprehensive genome- and transcriptome-wide analyses of mutations associated with MSI in Korean gastric cancer cell lines and primary tissues. We identified 18,377 MS mutations of five or more repeat nucleotides in coding sequences and untranslated regions of genes, and discovered 139 individual genes whose expression was down-regulated in association with UTR MS mutation. In addition, we found that 90.5% of MS mutations with deletions in gene regions occurred in UT...

Plasma Levels of Phospholipase A2-IIA in Patients with Different Types of Malignancies: Prognosis and Association with Inflammatory and Coagulation Biomarkers.
In conclusion, the study shows that the measurement of plasma sPLA2-IIA levels has prognostic values in patients with different types of malignancies. The association of sPLA2-IIA levels with CRP, fibrinogen, FVIII, and vWF levels supports the importance of inflammatory processes for the up-regulation of sPLA2-IIA during cancer progression. Environmental and lifestyle risk factors of gastric cancer.
Abstract Effective prevention and early diagnostic strategies are the most important public health interventions in gastric cancer, which remains a common malignancy worldwide. Preventive strategies require identification and understanding of environmental risk factors that lead to carcinogenesis. Helicobacter pylori (H. pylori) is the primary carcinogen as this ancient bacterium has a complex ability to interact with its human host. Smoking and salt are strong independent risk factors for gastric cancer whereas alcohol is only a risk when it is heavily consumed. Red meat and high fat increase the risk of gastric c...

Gastric Cancer Mortality in a High Incidence Area: Long-term follow-up of Helicobacter pylori-related Precancerous Lesions in the General Population.
CONCLUSIONS: In this population with a high rate of H. pylori infection, age over 50 years, smoking, family history of GC, IM, AG, and in particular, an undiagnosed GU were significant independent risk factors for mortality due to GC. The assessment of a combination of these risk factors might identify individuals at risk of GC who could possibly benefit from regular surveillance.
