BARCELONA — Allopurinol treatment for 6 weeks significantly improved symptoms of chronic, stable angina in a randomized, placebo-controlled, crossover study of 60 patients. Allopurinol administered at labeled dosages produced three significant improvements during an exercise treadmill test, which were the study's primary end points: increased total exercise time, increased time to ST-segment depression, and increased time to angina, Dr. Awsan Noman said at the annual congress of the European Society of Cardiology. CONCLUSION: Novel findings of our study suggest that lowering uric acid in ESRD patients may help to reduce the risk of cardiovascular disease in this population. It should be noted than an increase in triglycerides may mitigate the reduction in risk.

In this study, we used human astrocytoma 1321N1 cells to investigate the key molecules mediating sPLA2-IIA-induced cell proliferation. We found that sPLA2-IIA promoted reactive oxygen species (ROS) accumulation, which was abrogated in the presence of allopurinol and DPI, but not by rotenone, discarding mitochondria as a ROS source. In this study, we tested the sustainability of this enhanced contraction associated with decreased oxidative damage over an extended time. Mice were divided into three groups: sham-operated control, myocardial infarction-heart failure (MI-HF), and oxypurinol-treated heart failure (Oxy-HF). We demonstrated the development of antral ulcers induced in rats by alendronate and investigated the pathogenic factors involved in this model. Animals fasted for 18 h were given alendronate p.o., and then re-fed normally and killed on various days up to 7 days later. Alendronate caused non-hemorrhagic damage in both the corpus and antrum of fasted rats, but after refeeding for 3 days the lesions in the corpus healed completely, while those in the antrum developed into large ulcers with increased vascular permeability. Conclusion: A high fructose diet can induce metabolic syndrome even in the setting of caloric restriction. Captopril and allopurinol synergistically reduce features of the metabolic syndrome, especially hypertension, insulin resistance and dyslipidemia. Combination allopurinol and ACE inhibitor therapy might provide a superior means to prevent diabetes and cardiovascular disease.
