Dissolution behaviour of a poorly water-soluble drug, tadalafil, from its solid dispersion systems with poloxamer 407 has been investigated. Solid dispersion systems of tadalafil were prepared with poloxamer 407 in 1:0.5, 1:1.5 and 1:2.5 ratios using the melting method. Characterization of binary systems with FTIR and XRPD studies demonstrated the presence of strong hydrogen bonding interactions, a significant decrease in crystallinity and the possibility of existence of amorphous entities of the drug. The so-called PDE5 inhibitors are better at treating erectile dysfunction, according to a new guideline from the American College of Physicians.
Just in case anyone out there was unconvinced that erectile dysfunction is best treated by popping one of those well-advertised pills, the American College of Physicians has officially proclaimed the superiority of drugs such as Viagra, Levitra and Cialis over hormone treatment. Two closely related series of novel beta-carboline derivatives, electronically similar to tadalafil (CAS 171596-29-5), were synthesized and evaluated for their inhibitory effects upon phosphodiesterase 5 (PDE5) and phosphodiesterase 11 (PDE11) and their in vitro tumor cell growth inhibitory activity versus HT29 colorectal carcinoma cell line. Interestingly, some of the synthesized compounds showed growth inhibitory properties that appear to be associated with their ability to inhibit PDE5. Moreover, the PDE5 inhibition seems relevant to the stereochemical aspects of the compounds.

Flushing is one of the most common vasodilation-related adverse effects associated with both nitrates and phosphodiesterase type 5 (PDE5) inhibitors. The present study aimed to investigate the effects of orchidectomy and ovariectomy on isosorbide dinitrate-, sildenafil-, vardenafil- and tadalafil-induced flushing of tail-skin in mice. Background: The purpose of the study was to evaluate potency following Radical Retropubic Prostatectomy (RRP), in the era of phosphodiesterase (PDE5) inhibitors. Methods: 100 consecutive patients who underwent nerve sparing RRP and were potent preoperatively were analyzed postoperatively for the preservation of erectile function.
