Conclusions: The proportion of patients with anti-APC antibodies was low and was similar between DAA-treated and placebo-treated patients. No relationship between anti-APC antibody development and adverse reactions was observed. There was no evidence that the anti-APC antibodies detected represented a specific immune response to DAA therapy. (Source: Journal of Thrombosis and Haemostasis)

Conclusions: Socioeconomic inequalities in CHD persist in the elderly and are at least partly explained by behavioral risk factors; novel (inflammatory) coronary risk markers made some further contribution. Reducing inequalities in behavioral factors (especially cigarette smoking) could reduce these social inequalities by at least one-third. (Source: Journal of Thrombosis and Haemostasis)

This article represents an integrated summary of presentations given at the Fourth Annual Platelet Colloquium in January 2009. The process of and factors mediating platelet[ndash]platelet and platelet[ndash]leukocyte interactions in inflammatory and immune responses are discussed, with the roles of P-selectin, chemokines and Src family kinases being highlighted. Also discussed are specific disorders characterized by local or systemic platelet activation, including coronary artery restenosis after percutaneous intervention, alloantibody-mediated transplant rejection, wound healing, and heparin-induced thrombocytopenia. Summary. Venous thromboembolism (VTE) prevention has been recognized as the most important practice for improving patient safety in hospitals. To be effective, VTE prophylaxis must be appropriately prescribed with respect to type, dose and duration. Large-scale studies of medical discharge records have highlighted low rates of appropriate thromboprophylaxis in hospitalized medical patients, especially those with cancer or severe lung disease. Lack of prophylaxis and an insufficient duration are the most common forms of inappropriate prophylaxis. Multifaceted, active, quality improvement initiatives have been developed and ...

Summary. Background: Current guidelines state that platelet aggregation studies should be conducted within 4 h of venepuncture because of the decline in sensitivity to platelet agonists. This constrains studies of platelet activity in clinical situations where samples need to be transported or there are unavoidable delays prior to assessment. Objectives: The aim of the present study was to compare systematically the responses of platelets stored in the presence of either citrate or heparin, the two most widely used anti-coagulants, using a range of standard techniques. Methods: Blood was taken from healthy volunteers and e...

Conclusions: The decreased VWF:RCo seen with the P1467S sequence variation likely represents an artifact as a result of the use of ristocetin to measure VWF activity. Conclusions: Pre-hospital initiation of high-dose tirofiban reduces the 30-day incidence of stent thrombosis in STEMI patients treated with primary PCI and stenting. Early stent thrombosis and pre-hospital initiation of high-dose tirofiban were independent predictors of 30-day mortality. (Source: Journal of Thrombosis and Haemostasis)

Conclusions: SSL5 activates and aggregates platelets in a GPIb[alpha]-dependent manner, which could be important in colonization of the vascular bed and evasion of the immune system by S. aureus. (Source: Journal of Thrombosis and Haemostasis)

Conclusions: These data show that TM binding alters the conformation of thrombin in a similar manner as Na+ coordination, resulting in an ordering of the Na+ binding loop and an opening of the adjacent S1 pocket. We conclude that other, more subtle subsite changes are unlikely to influence thrombin specificity toward macromolecular substrates. (Source: Journal of Thrombosis and Haemostasis)

Evaluation of lyophilized platelets as an infusible hemostatic agent in experimental non-compressible hemorrhage in swine

Conclusions: This is the first reported study to evaluate rehydrated, lyophilized platelets as an infusible hemostatic agent for non-compressible hemorrhage. Stasix improved survival and reduced blood loss in a liver injury porcine model. However, evidence of thrombotic complications warrants further investigation prior to human use in the setting of traumatic hemorrhage. (Source: Journal of Thrombosis and Haemostasis)

Conclusions: The mutants showed no or marginally improved catalytic properties in FX activation by the intrinsic tenase complex (FIXa[ndash]FVIIIa[ndash]Ca2+[ndash]phospholipid). The combination of mutations Y94F and K98T hardly affected FX activation in the presence of FVIIIa, but yielded a FIX molecule that, in FIX-depleted plasma, had [sim] 2.5-fold higher clotting activity and [sim] 3.5-fold higher activity in a thrombin generation assay than plasma-derived FIX (pdFIX). Two FIXa mutants had considerably increased activities towards FX in the absence of FVIIIa. FIXa-Y94F/K98T/Y177F/I213V/E219G (FIXa-L) and FIXa-Y94F/A95...
