CYT997 Causes apoptosis in human multiple myelomaemail this articleEmail this article to a colleague. save this article to My ClippingsSave this article to My Clippings. discuss this articleDiscuss or comment on this article.
Summary  Multiple Myeloma (MM) is an incurable malignancy of mature plasma cells. Microtubule targeting agents (MTAs) are an established class of drug that include many conventional and some novel compounds. MTAs function by inhibiting the polymerisation or depolymerisation of microtubules (MTs) within the cell, disrupting various important cellular functions. We have investigated pre-clinically the novel tubulin polymerisation inhibitor CYT997 for the potential treatment of MM. Here we demonstrate the promising anti-myeloma activity of CYT997 as evidenced by tubulin disruption, inhibition of growth and proli...

Treatment of obstructive sleep apnea (OSA) in dentate patients using a mandibular advancement splint (MAS) from mandibular repositioners has been documented in detail. PPARγ Inhibitors as Novel Tubulin-Targeting Agentsemail this articleEmail this article to a colleague. save this article to My ClippingsSave this article to My Clippings. discuss this articleDiscuss or comment on this article.
The microtubule-targeting agents (MTAs) are a very successful class of cancer drugs with therapeutic benefits in both hematopoietic and solid tumors. However, resistance to these drugs is a significant problem. Current MTAs bind to microtubules, and/or to their constituent tubulin heterodimers, and affect microtubule polymerization and dynamics. The PPARγ inhibitor T0070907 can reduce tubulin levels in colorectal cancer cell lines and suppress tumor growth in a murine xenograft model. T0070907 does not alter microtubule polymerization in vitro, and does not appear to work by triggering modulation of tubulin RNA leve...

Subjects were eight healthy volunteers. Tongue pressure against the hard palate during swallowing 10 ml of water was measured under two conditions: preanesthesia and postanesthesia of anterior teeth. The sensory deprivation of anterior teeth was performed by periodontal anesthesia. Tongue...

Microtubules in apoptosis induction: are they necessary?email this articleEmail this article to a colleague. save this article to My ClippingsSave this article to My Clippings. discuss this articleDiscuss or comment on this article.
Microtubule-Targeting Agents (MTAs) constitute a class of drugs largely used in cancer treatment. Among them, both taxanes and Vinca-alkaloids are known to inhibit cancer cell proliferation by inducing cell cycle arrest and subsequent apoptosis. These agents modify the cytoskeleton by affecting the tubulin/microtubule system. In cancer cells, both classes suppress microtubule dynamics through inhibition of microtubule dynamic instability and treadmilling, and commonly induce diverse signals responsible for cell death initiation and execution via the mitochondrial intrinsic pathway. However, links between microtubule ne...

Anticancer agents that interfere with tubulin functions are widely used in the clinic and have a broad spectrum of activity against both haematological malignancies and solid tumours. Recently, MTAs have received considerable interest as potential tumour-selective anti-angiogenic and vascular-disrupting agents.
