Sodium butyrate-loaded nanoparticles coated with chitosan for the treatment of neovascularization in age-related macular degeneration: ocular biocompatibility and antiangiogenic activity
Eur J Pharm Biopharm. 2022 Aug 27:S0939-6411(22)00178-3. doi: 10.1016/j.ejpb.2022.08.011. Online ahead of print.ABSTRACTSodium butyrate-loaded nanoparticles coated chitosan (NaBu-loaded nanoparticles/CS) were developed to treat the choroidal neovascularization in wet age-related macular degeneration (AMD). The nanoparticles were produced by double emulsification and solvent evaporation technique, optimized by experimental statistical design, characterized by analytical methods, investigated in terms of in vitro and in vivo ocular biocompatibility, and evaluated as an antiangiogenic system in vivo. The NaBu-loaded nanoparti...

Effectiveness of Carbonic Anhydrase Inhibitor Loaded Nanoparticles in the Treatment of Diabetic Retinopathy
Biomed Phys Eng Express. 2023 Feb 9. doi: 10.1088/2057-1976/acba9d. Online ahead of print.ABSTRACTDiabetic Retinopathy (DRP) is a disease consisting of all the structural and functional changes that develop in the retinal layer of the eye due to diabetes. DRP is the most important cause of blindness between the ages of 20-74 in the world, and the most successful standard treatment option in the treatment of DRP is intravitreal injections. To synthesize acetazolamide loaded nanoparticles to be applied intravitreal treatment of DRP and to examine the in vitro efficacy of the nanoparticles. ACZ loaded PHBV nanoparticles (PHBV...

Sirolimus loaded chitosan functionalized poly (lactic-co-glycolic acid) (PLGA) nanoparticles for potential treatment of age-related macular degeneration
Int J Biol Macromol. 2021 Sep 15:S0141-8130(21)01991-7. doi: 10.1016/j.ijbiomac.2021.09.069. Online ahead of print.ABSTRACTThe usefulness of sirolimus (SIR) in the treatment of diseases that involve retinal degeneration like age-related macular degeneration (AMD) has been well documented. However, the problem still remains probably owing to the peculiar environment of the eye and/or unfavourable physiochemical profile of SIR. In the present work, we aimed to fabricate sirolimus loaded PLGA nanoparticles (SIR-PLGA-NP) and chitosan decorated PLGA nanoparticles (SIR-CH-PLGA-NP) to be administered via non-invasive subconjuncti...

Retinal cytotoxicity of silica and titanium dioxide nanoparticles
This study used R28 cells (a retinal precursor cell line that originated from rats) to investigate the safety of two commonly used types of NPs: silica nanoparticles (SiO2NPs, 100 nm) and titanium dioxide nanoparticles (TiO2NPs, 100 nm). Cellular viability and reactive oxygen species generation were measured after 24, 48, and 72 h of exposure to each NP. Cellular autophagy and the mTOR pathways were evaluated. The retinal toxicity of the NPs was investigated in vivo in rat models. Both types of NPs were found to induce significant dose-dependent toxicity on the R28 cells. A significant elevation of reactive oxygen species ...

Preparation, physicochemical, and retinal anti-angiogenic evaluation of poloxamer hydrogel containing dexamethasone/avastin-loaded chitosan-N-acetyl-L-cysteine nanoparticles
This study was meant to describe a Poloxamer hydrogel combining Chitosan-N-acetyl-L-cysteine (CNAC) nanoparticles to increase loading and sustained intravitreal administration of Avastin macromolecule. To increase the drug's efficacy and reduce the interfacial fluid pressure in a formulation, dexamethasone was used. To do so, CNAC was synthesized. Then, Avastin- loaded CNAC nanoparticles were prepared and optimized. The resulting hydrogel's sol-gel transition time and viscosity were determined using poloxamer and hydroxypropylmethylcellulose (HPMC). In vitro and in vivo investigations of Avastin-loaded CNAC nanoparticles a...

Post-self-repair process of neuron cells under the influence of neutral and cationic nanoparticles
Publication date: Available online 15 November 2019Source: Chinese Chemical LettersAuthor(s): Ting Wang, Guanwen Qu, Yu Deng, Jing Shang, Zhangqi Feng, Fengyu Yang, Nongyue He, Jie ZhengAbstractThe prevalence of functionalized nanoparticles in biological and clinical fields attracts intensive toxicology investigations. Minimizing the nanoparticles’ biohazard remains a challenge due to the insufficient understanding on the nanoparticle-induced cell death mechanism. In the presented study, we observed the lysosome and genome injuries and so caused cell cycle changes and regulations of retinal ganglion neuron cell 5 (RGC-5)...

Vitamin A- and C-rich Pouteria camito fru it derived superparamagnetic nanoparticles Synthesis, characterization, and their cytotoxicity
Conclusion: The Pouteria caimito fruit that possesses strong oxidizing and nontoxic properties can be a potentially attractive source for the production of iron oxide nanoparticles. Moreover, the cytotoxicity assay results revealed that iron oxide nanoparticles synthesized using the Pouteria caimito fruit extract derived can be used for targeting cancer cells and treating other diseases because of their nontoxic nature. These nanoparticles can be used for the treatment of cancer and other diseases in the future. Keywords: Fruit; Pouteria caimito; ferric chloride; superparamagnetic nanoparticles; cytotoxicity.

Cytotoxic effect of silica nanoparticles on human retinal pigment epithelial cells
In conclusion, SiO2 exposure can induce cytotoxic effects in RPE cell line. The results of this study provide clues that exposure to SiO2 nanoparticles may impair visual function and reduce quality of life. However, further studies are needed in this regard.PMID:37406486 | DOI:10.1016/j.bbrc.2023.06.083
