Topographic expression of the Hippo transducers TAZ and YAP in triple-negative breast cancer treated with neoadjuvant chemotherapy
The Hippo signaling acts as a tumor-suppressor pathway that negatively regulates TAZ and YAP. Increasing evidence supports the activation of TAZ and YAP in breast cancer. Moreover, the Hippo pathway is involve...

Evaluation of intraarterial and intravenous cisplatin chemotherapy in the treatment of metastatic osteosarcoma using an orthotopic xenograft mouse model
Osteosarcoma is the most common primary malignancy of bone. Its treatment relies on the administration of neoadjuvant and adjuvant chemotherapy combined with surgery. Alternative to common intravenous (i.v.) a...

The role of extracelluar matrix in osteosarcoma progression and metastasis
Osteosarcoma (OS) is the most common primary bone malignancy and responsible for considerable morbidity and mortality due to its high rates of pulmonary metastasis. Although neoadjuvant chemotherapy has improv...

Current treatment of rectal cancer: The watch-and-wait method. Are we there yet?
Abstract: Surgery remains the standard treatment of rectal carcinoma, with the use of neoadjuvant chemotherapy and radiation for locally advanced rectal carcinoma significantly decreasing local recurrence rates, as well as providing tumor downstaging. In those patients who exhibit complete pathological response, the question of the benefit of surgical resection and the utility of a ‘wait-and-see’ approach has been raised. However, the variability in determining a true pathological complete response, as well as the inability to accurately stage the nodes, argues against a non-operative approach. Many small series have a...

Are Females Predisposed to Complications in Head and Neck Cancer Free Flap Reconstruction?
Conclusions: Multivariate analysis of the data revealed that the factor associated with an increased incidence of postoperative complications was preoperative radiotherapy. Moreover, the variable that remained statistically significant and correlated with an increased incidence and severity of free flap reconstruction complications was female gender. Identifying the etiology of these findings is an intriguing field for additional research.

Incidence and location of lymph node metastases in patients undergoing radical cystectomy for clinical non–muscle invasive bladder cancer: Results from a prospective lymph node mapping study
Conclusions: Routine LND is important in patients with cT1 and cTis-only bladder cancer, but may have limited value in patients with cTa. LNM beyond the boundaries of a standard LND occurred in up to one-third of node-positive patients. In the absence of skip metastases, however, performing a standard LND would correctly identify all node-positive patients. Whether removal of LNM proximal to the common iliac vessels provides a survival benefit remains to be evaluated in future prospective studies.

The presence of positive surgical margins in patients with organ-confined prostate cancer results in biochemical recurrence at a similar rate to that in patients with extracapsular extension and PSA≤10ng/ml
Abstract: Purposes: We investigated whether patients with organ-confined prostate cancer (PCa) and positive surgical margins (SMs) had a similar biochemical recurrence (BCR) risk compared with patients with pT3a and preoperative prostate-specific antigen (PSA) levels≤10ng/ml. Furthermore, we examined the effects of incorporating SM status, Gleason score (Gls), and preoperative PSA level into the discrimination accuracy of the current tumor node metastasis-staging system.Materials and methods: We analyzed 863 PCa patients treated with radical prostatectomy from 1999 to 2008. Only individuals with pT2N0 or pT3N0, without n...

High rates of advanced disease, complications, and decline of renal function after radical nephroureterectomy
Abstract: Objectives: Recurrences remain common following radical nephroureterectomy (RNU) for locally advanced upper-tract urothelial carcinoma (UTUC). We review a cohort of RNU patients to identify the incidence of locally advanced disease, decline in renal function, complications, and utilization of adjuvant chemotherapy (AC).Methods: Institutional databases from 7 academic medical centers identified 414 RNU patients treated between 2003 and 2012 who had not received neoadjuvant chemotherapy. Glomerular filtration rate was estimated using the Modification of Diet in Renal Disease equation. Complications were classified ...

Effect of diabetes mellitus and metformin use on oncologic outcomes of patients treated with radical cystectomy for urothelial carcinoma
Conclusions: Diabetic patients who do not use metformin appear to be at higher risk of cancer-specific and any-cause mortality than patients without DM. It remains unclear, whether the severity of DM in this group of patients or the use of metformin itself affects outcomes of UCB. The mechanisms behind the effect of DM on patients with UCB and the potential protective effect of metformin need further elucidation.

Abstract PL01-01: Distinct immune and metabolic signatures in prostate and breast tumors of African American patients
Not all segments of the U.S. population have equally benefited from the advances in our knowledge and treatment of cancer. As a result, some population groups experience a disproportionally high cancer burden, leading to excessively high cancer mortality rates in the African American community when compared with other communities. For example, African American patients have the highest prostate and breast cancer mortalities among all U.S. racial and ethnic groups. This survival disparity has been attributed to a number of factors including differences in insurance status and medical care. However, other observations sugges...

Dual HER2 blockade in the neoadjuvant and adjuvant treatment of HER2-positive breast cancer
Pooja Advani,1 Lauren Cornell,2 Saranya Chumsri,1 Alvaro Moreno-Aspitia1 1Division of Hematology and Oncology, 2Department of Internal Medicine, Mayo Clinic, Jacksonville, FL, USA Abstract: Human epidermal growth factor receptor 2 (HER2) is a tyrosine kinase transmembrane receptor that is overexpressed on the surface of 15%–20% of breast tumors and has been associated with poor prognosis. Consistently improved pathologic response and survival rates have been demonstrated with use of trastuzumab in combination with standard chemotherapy in both early and advanced breast cancer. However, resistance to trastuzumab may p...
