Primary melanoma of female urethra: A case diagnosed in Ouagadougou
We report a case of primary melanoma of a female urethra diagnosed at a non-metastatic stage in a 48-year-old patient with a history of breast carcinoma treated with radiotherapy and hormone therapy. The patient was consulting for dysuria, hematuria, and perineal pain. The clinical examination found a prolapsed and black mass, developed at the expense of the urethra and located at the anterosuperior part of the vulva. The mass biopsy revealed a proliferation of fusiform and globular cells loaded with black pigment expressing the anti-HMB 45 and PS 100 antibodies. The extension assessment showed an absence of secondary loca...

NAT10 regulates the repair of UVB-induced DNA damage and tumorigenicity
Toxicol Appl Pharmacol. 2023 Sep 14:116688. doi: 10.1016/j.taap.2023.116688. Online ahead of print.ABSTRACTChemical modifications in messenger RNA (mRNA) regulate gene expression and play critical roles in stress responses and diseases. Recently we have shown that N6-methyladenosine (m6A), the most abundant mRNA modification, promotes the repair of UVB-induced DNA damage by regulating global genome nucleotide excision repair (GG-NER). However, the roles of other mRNA modifications in the UVB-induced damage response remain understudied. N4-acetylcytidine (ac4C) is deposited in mRNA by the RNA-binding acetyltransferase NAT10...

Nobiletin regulates invasion, migration and metastasis in hypopharyngeal squamous cell carcinoma
This study examined the effects of NOB on the proliferation, migration, and invasiveness of the FaDu cell line derived from hypopharyngeal squamous cell carcinoma. We determined protein kinase phosphorylation by western blot analysis, migration by Transwell assay, and metastatic potential by enzyme-linked immunosorbent assay of vascular endothelial growth factor. NOB inhibited cell proliferation by 40% at concentrations of 15 µM and 40 µM, and reduced migration and induced apoptosis at 50 µM by downregulating extracellular signal-regulated kinase, protein kinase B, and phosphoinositide 3-kinase. Our results suggest that...

Molecular Subtypes Based on PANoptosis Genes and Characteristics of Immune Infiltration in Cutaneous Melanoma
Cell Mol Biol (Noisy-le-grand). 2023 Aug 31;69(8):1-8. doi: 10.14715/cmb/2023.69.8.1.ABSTRACTCutaneous melanoma (CM) remains the most life-threatening form of skin cancer. Further risk stratification and search for new prognostic targets for CM are of positive clinical significance. PANoptosis is defined as an inflammatory programmed cell death mediated by the PANoptosome complex and cannot be characterized by pyroptosis, apoptosis or necroptosis alone. Although PANoptosis is closely associated with many diseases including cancer, it has not been reported in CM. Combined with GTEx and TCGA database, we extracted 14 PANopto...

Targeted proteomic quantitation of NRF2 signaling and predictive biomarkers in HNSCC
Mol Cell Proteomics. 2023 Sep 14:100647. doi: 10.1016/j.mcpro.2023.100647. Online ahead of print.ABSTRACTThe NFE2L2(NRF2) oncogene and transcription factor drives a gene expression program that promotes cancer progression, metabolic reprogramming, immune evasion and chemoradiation resistance. Patient stratification by NRF2 activity may guide treatment decisions to improve outcome. Here, we developed a mass spectrometry (MS)-based targeted proteomics assay based on internal standard triggered parallel reaction monitoring (IS-PRM) to quantify 69 NRF2 pathway components and targets, as well as 21 proteins of broad clinical si...

A Subungual Nevoid Melanoma of the Right Hallux from a 65-Year-Old Man: A Case Report
J Am Podiatr Med Assoc. 2023 Jul-Aug;113(4):21-108. doi: 10.7547/21-108.ABSTRACTNevoid melanoma (NeM) is a rare variant of malignant melanoma characterized by slight cellular atypia, polymorphism, and incomplete maturation. It most frequently occurs on the trunk and arms but rarely on the foot. Here, we report a subungual NeM of the right hallux. A 65-year-old man presented with severe pain of 6 months' duration to his right great toe following self-treatment for an ingrown nail. He was evaluated and treated with debridement of the toenail at an urgent medical center 3 months prior. However, this had not relieved his pain....

Porocarcinoma of the Left Foot: A Case Review
We present the case of a 62-year-old man with a lesion on the left foot. The diagnosis of the initial biopsy was squamous cell carcinoma. Six months later, the lesion reoccurred, and a second biopsy confirmed it to be eccrine porocarcinoma.PMID:37715975 | DOI:10.7547/17-069

Cancer cell-intrinsic PD-1: Its role in malignant progression and immunotherapy
Biomed Pharmacother. 2023 Sep 14;167:115514. doi: 10.1016/j.biopha.2023.115514. Online ahead of print.ABSTRACTProgrammed cell death protein-1 (PD-1), also called CD279, is coded by the PDCD1 gene and is constitutively expressed on the surface of immune cells. As a receptor and immune checkpoint, PD-1 can bind to programmed death ligand-1/programmed death ligand-2 (PD-L1/PD-L2) in tumor cells, leading to tumor immune evasion. Anti-PD-1 and anti-PD-L1 are important components in tumor immune therapy. PD-1 is also expressed as an intrinsic variant (iPD-1) in cancer cells where it plays important roles in malignant progression...
