Prevalence of mutations in common tumour types in Northern England and comparable utility of national and international Trial Finders
CONCLUSIONS: Regional differences in mutations may exist with advanced stage accounting for prevalence of specific mutations. A national Trial Finder shows utility in finding targeted trials whilst commercial sequencing reports may over-report 'actionable' mutations. Understanding local prevalence and trial availability could increase enrolment onto matched early phase trials.PMID:37702806 | DOI:10.1007/s00432-023-05365-y

Battling colorectal cancer via s-triazine-based MMP-10/13 inhibitors armed with electrophilic warheads for concomitant ferroptosis induction; the first-in-class dual-acting agents
Bioorg Chem. 2023 Sep 7;141:106839. doi: 10.1016/j.bioorg.2023.106839. Online ahead of print.ABSTRACTThere is an increasing interest in halting CRC by combining ferroptosis with other forms of tumor cell death. However, ferroptosis induction is seldom studied in tandem with inhibiting MMPs. A combination that is expected to enhance the therapeutic outcome based on mechanistic ferroptosis studies highlighting the interplay with MMPs, especially MMP-13 associated with CRC metastasis and poor prognosis. Herein, we report new hybrid triazines capable of simultaneous MMP-10/13 inhibition and ferroptosis induction bridging the g...

Activation of vascular endothelial cells by synovial fibrosis promotes Netrin-1-induced sensory nerve sprouting and exacerbates pain sensitivity
J Cell Mol Med. 2023 Sep 13. doi: 10.1111/jcmm.17950. Online ahead of print.ABSTRACTSynovial fibrosis is one of the most dominant histopathological changes in osteoarthritis of the knee (KOA), and activation of vascular endothelial cells in synovial fibrosis is both an important factor in mediating pain in KOA and a major contributor to the generation of pain signals. At the same time, angiogenesis and nerve fibres are more likely to underlie the pathology of pain induced by synovial fibrosis. In the present study, we established a co-culture model of human umbilical vein endothelial cells (HUVECs) with dorsal root ganglio...

Different oncological features of colorectal cancer codon-specific < em > KRAS < /em > mutations: Not codon 13 but codon 12 have prognostic value
CONCLUSION: This study provides evidence that KRAS codon 13 mutation is less likely to serve as a prognostic biomarker than codon 12 mutation for CRC in a large-scale cohort.PMID:37701134 | PMC:PMC10494767 | DOI:10.3748/wjg.v29.i32.4883

MicroRNAs Regulate Tumorigenesis by Downregulating SOCS3 Expression: An < em > In silico < /em > Approach
This study identifies the interactions of known (miR-203a-5p) and novel (miR-6756-5p, miR-6732-5p, miR-1203, miR-6887-5p) microRNAs with SOCS3 regions due to their maximum interactions. Identifying the interactions of these microRNAs with SOCS3 will significantly advance the understanding of oncomiRs (miRNAs that are associated with cancer development) in tumor development due to their influence on SOCS3 expression. These insights will assist in future studies to understand the significance of miRNA-SOCS3-associated tumor development and progression.PMID:37701630 | PMC:PMC10493049 | DOI:10.1177/11779322231193535

Generalizability of Self-Supervised Training Models for Digital Pathology: A Multicountry Comparison in Colorectal Cancer
CONCLUSION: We find that using unlabeled image data for SSL pretraining in CRC related tasks is more effective than using ImageNet pretraining. Furthermore, SSL pretraining using data from multiple countries achieve more stable performance and better generalization than single-country pretraining.PMID:37703507 | DOI:10.1200/CCI.22.00178
