Loss of endothelial glucocorticoid receptor accelerates organ fibrosis in < em > db/db < /em > mice
This study aimed to determine the effect of loss of endothelial GR on organ fibrosis in the db/db model. Db/Db mice lacking endothelial GR showed more severe fibrosis in multiple organs compared to endothelial GR-replete db/db mice. Organ fibrosis could be substantially improved either through administration of a Wnt inhibitor or metformin. IL-6 is a key cytokine driving the fibrosis phenotype and is mechanistically linked to Wnt signaling. The db/db model is an important tool to study mechanisms of fibrosis and its phenotype in the absence of endothelial GR highlights the synergistic effects of Wnt signaling and inflammat...

Metformin derivatives - Researchers' Friends or Foes?
Biochem Pharmacol. 2023 Aug 15:115743. doi: 10.1016/j.bcp.2023.115743. Online ahead of print.ABSTRACTMetformin has been used for ages to treat diabetes mellitus due to its safety profile and low cost. However, metformin has variable pharmacokinetics in patients, and due to its poor oral absorption, the therapeutic doses are relatively high, causing unpleasant gastrointestinal adverse effects. Therefore, novel derivatives of metformin have been synthesized during the past decades. Particularly, after the mid-2000s, when organic cation transporters were identified as the main metformin carriers, metformin derivatives have be...
