Obtaining a high titer of polyclonal antibodies from rats to the SARS-CoV-2 nucleocapsid protein and its N- and C-terminal domains for diagnostic test development
J Immunol Methods. 2023 Sep 11:113558. doi: 10.1016/j.jim.2023.113558. Online ahead of print.ABSTRACTSevere Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is an enveloped, plus-stranded RNA virus responsible for the Coronavirus Disease 2019 (COVID-19). Patients infected with COVID-19 may be asymptomatic or have symptoms ranging from mild manifestations to severe cases of the disease that could lead to death. The SARS-CoV-2 genome encodes 4 structural proteins, including the Spike protein (S), the Nucleocapsid protein (N), membrane protein (M) and, the envelope (E). The N protein forms a major component of the ribonu...

Pharma - manufacturing: the unappreciated and overlooked indispensable skill
Curr Opin Immunol. 2023 Sep 11;84:102385. doi: 10.1016/j.coi.2023.102385. Online ahead of print.ABSTRACTThe process of vaccine production, manufacturing, is time-intensive, complex, expensive, and highly technical, requiring close coordination and collaboration among multiple companies with different inputs, from active pharmaceutical ingredients to glass, and specializations, and with the supply chains spread across many countries. Covid-19 pandemic highlighted that neglecting and ignoring the need for a global effort in vaccine manufacturing and delivery can have alarming, and devastating, repercussions, especially when ...

Effects of Skeleton Structure of Mesoporous Silica Nanoadjuvants on Cancer Immunotherapy
Three commonly used mesoporous silica nanoparticles (MSNs) with different skeletons are synthesized by using different silica sources, such as TEOS for MSN, TEOS/BTEE for EMSN, and TEOS/BTES for TMSN. Compared with other two OMSNs, the OTMSN effectively targeted lymph nodes, activated DCs maturation, stimulated the proliferation of CD4+, CD8+ T cells, and polarized M2 phenotype macrophages into M1 phenotype macrophages, and reversed immunosuppressive tumor microenvironment. AbstractMesoporous silica nanoparticles (MSNs) have been widely praised as nanoadjuvants in vaccine/tumor immunotherapy thanks to their excellent bioco...

Emergence of Marburg virus: a global perspective on fatal outbreaks and clinical challenges
The Marburg virus (MV), identified in 1967, has caused deadly outbreaks worldwide, the mortality rate of Marburg virus disease (MVD) varies depending on the outbreak and virus strain, but the average case fatality rate is around 50%. However, case fatality rates have varied from 24 to 88% in past outbreaks depending on virus strain and case management. Designated a priority pathogen by the National Institute of Allergy and Infectious Diseases (NIAID), MV induces hemorrhagic fever, organ failure, and coagulation issues in both humans and non-human primates. This review presents an extensive exploration of MVD outbreak evolu...

Burdening Perspectives and Treatment Modalities of Monkeypox: A Central Dogma
Infect Disord Drug Targets. 2023 Aug 25. doi: 10.2174/1871526523666230825164222. Online ahead of print.ABSTRACTThe monkeypox virus (MPXV), belonging to the genus Orthopoxvirus, is responsible for causing the zoonotic illness known as Monkeypox. The virus was initially identified during an outbreak at a Danish Zoo in 1958 and has since been found to infect various mammal species worldwide. While African squirrels and other rodents are believed to be the primary hosts, determining the natural host has proven challenging. While MPXV can be studied using different animal models in laboratory settings, understanding its natural...

Enhanced systemic tumor suppression by in situ vaccine combining radiation and OX40 agonist with CpG therapy
CONCLUSION: This triple combining in situ vaccine induced intensive antitumor responses, mediated effective systemic tumor control and survival benefit, and displayed impressive synergistic antitumor effect with checkpoint blockade therapy. These data preliminary confirmed the efficacy, feasibility and safety of the triple combining in situ vaccine, suggesting its great application potential as both monotherapy and a part of combined immunotherapeutic regimens in clinical scenario.PMID:37700338 | DOI:10.1186/s12967-023-04504-w

Macrophage colony-stimulating factor as a weapon against cytomegalovirus
EMBO Mol Med. 2023 Sep 12:e18319. doi: 10.15252/emmm.202318319. Online ahead of print.ABSTRACTCytomegalovirus (CMV) infection is one of the severe opportunistic infections faced by severely immunocompromised patients. High viral loads cause tissue-invasive disease and expose to death or various indirect effects. Substantial progress was made in monitoring active infection, and antiviral drugs were developed. However, dose-limiting toxicities and genotypic resistance limit therapeutic efficacy and vaccine development is hampered by the complex biology of the virus. In this issue of EMBO Molecular Medicine, Kandalla et al (2...
