Leveraging decagonal in-silico strategies for uncovering IL-6 inhibitors with precision
Comput Biol Med. 2023 Jul 3;163:107231. doi: 10.1016/j.compbiomed.2023.107231. Online ahead of print.ABSTRACTInterleukin-6 upregulation leads to various acute phase reactions such as local inflammation and systemic inflammation in many diseases like cancer, multiple sclerosis, rheumatoid arthritis, anemia, and Alzheimer's disease stimulating JAK/STAT3, Ras/MAPK, PI3K-PKB/Akt pathogenic pathways. Since no small molecules are available in the market against IL-6 till now, we have designed a class of small bioactive 1,3 - indanedione (IDC) molecules for inhibiting IL-6 using a decagonal approach computational studies. The IL-...

Determination of methotrexate in plasma and environmental samples using an electrochemical sensor modified by UiO66-NH < sub > 2 < /sub > /mesoporous carbon nitride composite and synergistic signal amplification with decorated AuNPs
In this study, UiO66-NH2/mesoporous carbon nitride (M - C3N4) composite provides a porous structure for decorated Au nanoparticles (AuNPs) to prepare a potent modifier for carbon paste electrode (CPE). Due to environmental toxicity of methotrexate, its sensitive, fast and, low-cost determination in workplace environments is of great interest. So, the modified CPE was applied as a sensitivity analysis approach for methotrexate in plasma samples. Cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were used as techniques to optimize the analysis and measurement of methotrexate. To measure this drug, several effe...

Compared efficacy of rituximab, abatacept, and tocilizumab in patients with rheumatoid arthritis refractory to methotrexate or TNF inhibitors agents: a systematic review and network meta-analysis
CONCLUSION: We found high heterogeneity among studies comparing rituximab, abatacept, and tocilizumab. On multivariate metaregressions, if the conditions of the RCTs were similar, we estimate that abatacept could increase the chance of reaching an ACR70 response by 2.2-fold compared to tocilizumab.PMID:37415193 | DOI:10.1186/s42358-023-00298-z

Takayasu Arteritis: A Difficult Diagnosis in a Patient With an Extensive Cardiovascular History
We present a case of a 57-year-old male with a history of extensive cardiovascular disease, initially attributed to atherosclerosis, resulting in several interventions, such as catheterization and major cardiac surgery, which didn't help improve his symptoms. Further evaluation revealed diffuse wall thickening of the aorta and its roots, as well as labs that suggested elevated inflammatory markers, comprehensive review of his chart and previous admissions, revealed that he had a well-documented aortitis for which he underwent a biopsy, which at the time was unrevealing. Furthermore, as he had significant aortic aneurysmal ...

Methotrexate degradation in artificial wastewater using non-thermal pencil plasma jet
Environ Sci Pollut Res Int. 2023 Jul 3. doi: 10.1007/s11356-023-28502-z. Online ahead of print.ABSTRACTThe rising global cancer rate is driving up the consumption of anticancer drugs. This causing a noticeable increase in the levels of these drugs in wastewater. The drugs are not metabolized effectively by the human body, leading to their presence in human waste, as well as in the effluent from hospitals and drug manufacturing industries. Methotrexate is a commonly used drug for treating various types of cancer. Its complex organic structure makes it difficult to degrade using conventional methods. The present work propose...

Immunosuppressive nanoparticles containing recombinant PD-L1 and methotrexate alleviate multi-organ inflammation
In this study, recombinant murine PD-L1 (rmPD-L1) was used for controlling T cell immunity to treat multi-organ inflammation. To enhance the immunosuppressive effect, we incorporated methotrexate, an anti-inflammatory drug, into hybrid nanoparticles (HNPs) and decorated the surface of HNPs with rmPD-L1 to produce immunosuppressive HNPs (IsHNPs). IsHNP treatment effectively targeted PD-1-expressing CD4 and CD8 T cells in the splenocytes; additionally, it promoted the production of Foxp3-expressing regulatory T cells, which suppressed the differentiation of helper T cells. IsHNP treatment also inhibited anti-CD3 antibody-med...
