Allosteric switch for electrochemical aptasensor toward heavy metals pollution of Lentinus edodes sensitized with porphyrinic metal-organic frameworks
We present an appropriate electrochemical aptasensor for Ag + assay based on biomimetic catalysis of porphyrin-encapsulated MOF (PorMOF) and allosteric switch of C-rich DNA. Thanks to the excellent biocompatibility, PorMOFs as nanozyme are used to design signal probes by loading duplex-like DNA scaffold. Owing to the specific recognition of Ag+ toward cytosine (C) base-rich DNA, PorMOF at the distal end was close to the underlying electrode via C-Ag+-C formation, leading to an enhanced current of catalytic hydroxylamine oxidation for signal generation. Using the positive correlation between current response and Ag+ level, ...

Mitigation of Anti ‐Drug Antibody Production for Augmenting Anticancer Efficacy of Therapeutic Protein via Co‐Injection of Nano‐Rapamycin
Co-injection of the PEGylated rapamycin nanoparticles and a protein toxin (trichosanthin) can mitigate the formation of anti-drug antibody via inducing durable antigen-specific tolerance. The combination of trichosanthin and the rapamycin nanoparticles yields an enhanced effect to inhibit the growth of breast tumor. AbstractThe induction of anti-drug antibody (ADA) is a formidable challenge for protein-based therapy. Trichosanthin (TCS) as a class of ribosome-inactivating proteins is widely studied in tumor treatment. However, the immunogenicity can induce the formation of ADA, which can cause hypersensitivity reactions an...

Expanding the ubiquitin code in pancreatic cancer
Biochim Biophys Acta Mol Basis Dis. 2023 Sep 11:166884. doi: 10.1016/j.bbadis.2023.166884. Online ahead of print.ABSTRACTThe ubiquitin-proteasome system (UPS) is a fundamental regulatory mechanism in cells, vital for maintaining cellular homeostasis, compiling signaling transduction, and determining cell fates. These biological processes require the coordinated signal cascades of UPS members, including ubiquitin ligases, ubiquitin-conjugating enzymes, deubiquitinases, and proteasomes, to ubiquitination and de-ubiquitination on substrates. Recent studies indicate that ubiquitination code rewriting is particularly prominent ...

Alkaloids as Additional Weapons in the Fight against Breast Cancer: A review
This study also includes an overview of synthesized analogues and pharmacological characteristics that will be beneficial to scientists working on alkaloids for medicinal purposes. In a recent survey of the literature, alkaloids are an important component of plant-derived antitumor medicines that hold great potential for the future development of cancer therapy and preventive therapies. We have also discussed structural analysis relationship (SAR) studies. Moreover, it covers clinical trial medications and FDA-approved medicines from the last five years that will be useful in further research.PMID:37702171 | DOI:10.2174/09...

Peroxisome proliferator-activated receptors as therapeutic target for cancer
J Cell Mol Med. 2023 Sep 12. doi: 10.1111/jcmm.17931. Online ahead of print.ABSTRACTPeroxisome proliferator-activated receptors (PPARs) are transcription factors belonging to the nuclear receptor family. There are three subtypes of PPARs, including PPAR-α, PPAR-β/δ and PPAR-γ. They are expressed in different tissues and act by regulating the expression of target genes in the form of binding to ligands. Various subtypes of PPAR have been shown to have significant roles in a wide range of biological processes including lipid metabolism, body energy homeostasis, cell proliferation and differentiation, bone formation, tiss...

Lnc-ing epigenetic mechanisms with autophagy and cancer drug resistance
Adv Cancer Res. 2023;160:133-203. doi: 10.1016/bs.acr.2023.03.002. Epub 2023 Apr 7.ABSTRACTLong noncoding RNAs (lncRNAs) comprise a diverse class of RNA molecules that regulate various physiological processes and have been reported to be involved in several human pathologies ranging from neurodegenerative disease to cancer. Therapeutic resistance is a major hurdle for cancer treatment. Over the past decade, several studies has emerged on the role of lncRNAs in cancer drug resistance and many trials have been conducted employing them. LncRNAs also regulate different cell death pathways thereby maintaining a fine balance of ...

PFKP: More than phosphofructokinase
Adv Cancer Res. 2023;160:1-15. doi: 10.1016/bs.acr.2023.03.001. Epub 2023 Mar 30.ABSTRACTPhosphofructokinase (PFK) is one of the key enzymes that functions in glycolysis. Studies show that PFKP regulates cell proliferation, apoptosis, autophagy, cell migration/metastasis, and stemness through glycolysis and glycolysis-independent functions. PFKP performs its function not only in the cytoplasm, but also at the cell membrane, on the mitochondria, at the lysosomal membrane, and in the nucleus. The functions of PFKP are extensively studied in cancer cells. PFKP is also highly expressed in certain immune cells; nevertheless, th...
