The Role of Endoplasmic Reticulum in Lipotoxicity during MASLD Pathogenesis
Am J Pathol. 2023 Sep 7:S0002-9440(23)00314-0. doi: 10.1016/j.ajpath.2023.08.007. Online ahead of print.ABSTRACTPerturbations in lipid and protein homeostasis induce endoplasmic reticulum (ER) stress in metabolic-dysfunction-associated steatotic liver disease (MASLD), formerly known as nonalcoholic fatty liver disease (NAFLD). Lipotoxic and proteotoxic stress can activate the unfolded protein response (UPR) transducers: Inositol Requiring Enzyme1α, PKR-like ER Kinase and Activating Transcription Factor 6α. Collectively, these pathways induce expression of genes that encode functions to resolve the protein folding defect ...

Chemical Synthesis and Biochemical Properties of Cholestane-5 α,6β-diol-3-sulfonate, A Non-hydrolysable Analogue of cholestane-5α,6β-diol-3β-sulfate
J Steroid Biochem Mol Biol. 2023 Sep 6:106396. doi: 10.1016/j.jsbmb.2023.106396. Online ahead of print.ABSTRACTCholestane-3β,5α,6β-triol (CT) is a primary metabolite of 5,6-epoxycholesterols (5,6-EC) that is catalyzed by the cholesterol-5,6-epoxide hydrolase (ChEH). CT is a well-known biomarker for Niemann-Pick disease type C (NP-C), a progressive inherited neurodegenerative disease. On the other hand, CT is known to be metabolized by the 11β-hydroxysteroid-dehydrogenase of type 2 (11β-HSD2) into a tumor promoter named oncosterone that stimulates the growth of breast cancer tumors. Sulfation is a major metabolic trans...

Association Between Whole Blood Selenium Levels and Triglyceride-to-High-Density Lipoprotein Cholesterol Ratio Among the General Population
Biol Trace Elem Res. 2023 Sep 9. doi: 10.1007/s12011-023-03839-1. Online ahead of print.ABSTRACTOur study aimed to examine whether whole blood selenium (WBSe) levels are related to the triglyceride-to-high-density lipoprotein cholesterol (TG/HDL-C) ratio among the general population. A total of 13,470 adults were included and analyzed from the National Health and Nutrition Examination Survey (NHANES) 2011-2018. In multivariable analyses, LnWBSe levels were significantly related to Ln(TG/HDL-C) ratio in fully adjusted model (β = 0.35; 95% confidence interval (CI): 0.22, 0.48; P < 0.001). Furthermore, the highest quartil...

Proprotein convertase subtilisin/kexin 9 (PCSK9) in patients with diffuse systemic sclerosis: A marker of disease activity and severe disease manifestations with potential therapeutic implementations
CONCLUSION: Serum PCSK9 is upregulated in d-SSc with higher levels in severe disease manifestations such as DU and ILD. It is correlated well with disease activity, more severe disease manifestations, and CIMT. The PCSK9 inhibitors may be a target of therapy in diseases with premature atherosclerosis such as d-SSc regardless of its anti-cholesterol effect, at least in more severe manifestations.PMID:37680510 | PMC:PMC10481689 | DOI:10.46497/ArchRheumatol.2023.9638

Usp25-Erlin1/2 activity limits cholesterol flux to restrict virus infection
Dev Cell. 2023 Aug 28:S1534-5807(23)00409-4. doi: 10.1016/j.devcel.2023.08.013. Online ahead of print.ABSTRACTReprogramming lipid metabolic pathways is a critical feature of activating immune responses to infection. However, how these reconfigurations occur is poorly understood. Our previous screen to identify cellular deubiquitylases (DUBs) activated during influenza virus infection revealed Usp25 as a prominent hit. Here, we show that Usp25-deleted human lung epithelial A549 cells display a >10-fold increase in pathogenic influenza virus production, which was rescued upon reconstitution with the wild type but not the ...

Ginsenoside Rb1 alleviates chronic intermittent hypoxia-induced diabetic cardiomyopathy in db/db mice by regulating the adenosine monophosphate-activated protein kinase/Nrf2/heme oxygenase-1 signaling pathway
CONCLUSION: Rb1 treatment alleviated CIH-induced diabetic cardiomyopathy and glycolipid metabolism disorders in db/db mice by inhibiting oxidative stress and regulating the AMPK/Nrf2/HO-1 signaling pathway.PMID:37679978 | PMC:PMC10465847 | DOI:10.19852/j.cnki.jtcm.20221206.004

Broad-Spectrum Antiviral Effect of Cannabidiol Against Enveloped and Nonenveloped Viruses
Conclusions: It was demonstrated that CBD inhibits structurally dissimilar viruses, suggesting that this phytochemical has broad-spectrum antiviral effect, representing a valuable alternative in emergency situations during viral outbreaks, like the one caused by severe acute respiratory syndrome coronavirus 2 in 2020.PMID:37682578 | DOI:10.1089/can.2023.0103

Adherence to aerobic training combined with high protein intake is associated with low blood pressure in Italian older adults: a cross-sectional study
CONCLUSIONS: Our results indicate that AT combined with HPI was negatively associated with SBP in a large and relatively unselected sample of Italian older adults living in the community. These findings need confirmation by ad hoc designed studies.PMID:37682490 | DOI:10.1007/s40520-023-02549-x
