MicroRNAs and their regulators: Potential therapeutic targets in pulmonary arterial hypertension
Vascul Pharmacol. 2023 Sep 10;153:107216. doi: 10.1016/j.vph.2023.107216. Online ahead of print.ABSTRACTPulmonary arterial hypertension (PAH) is a complex and progressive disease characterized by pulmonary arterial remodeling. Despite that current combination therapy has shown improvement in morbidity and mortality, a better deciphering of the underlying pathological mechanisms and novel therapeutic targets is urgently needed to combat PAH. MicroRNA, the critical element in post-transcription mechanisms, mediates cellular functions mainly by tuning downstream target gene expression. Meanwhile, upstream regulators can regul...

Adipocyte-derived small extracellular vesicles (ad-sEVs) have been implicated as a mecha-nism in several obesity-related diseases. We hypothesized that ad-sEVs from patients with AD would contain miRNAs predicted to downre...

GSE239453 Casein kinase 1 and 2 phosphorylate Argonaute proteins to regulate miRNA-mediated gene silencing
We report here that in C. elegans, two conserved serine/threonine kinases - Casein Kinase 1 alpha 1 (CK1A1) and Casein Kinase 2 (CK2) - regulate a highly conserved phosphorylation cluster of 4 Serine residues (S988:S998) on the miRNA-specific AGO protein ALG-1. We show that CK1A1 phosphorylates ALG-1 at sites S992 and S995, while CK2 phosphorylates ALG-1 at sites S988 and S998. Furthermore, we demonstrate that phospho-mimicking mutants of the entire S988:S998 cluster rescue the various developmental defects observed upon depleting CK1A1 and CK2. In humans, we show that CK1A1 also acts as a priming kinase of this cluster on...

Expression of microRNA-379 reduces metastatic spread of prostate cancer
DiscussionTaken together, our in vitro and in vivo functional assays support a role for miR-379 as a tumour suppressor. A potential mechanism is unravelled whereby miR-379 deregulation in PCa cells affects the secretion of GDF-15 from osteoblasts which in turn facilitates the metastatic establishment in bone. Our findings support the potential role of miR-379 as a therapeutic solution for prostate cancer.

Specific downregulation of microRNA-186 induces neural stem cell self-renewal by upregulating Bmi-1/FoxG1 expression
In this study, we isolated primary NSCs from the forebrain of fetal rat for in vitro analysis. Downregulation of miRNA-186 in response to a specific miRNA inhibitor resulted in upregulation of Bmi-1 and FoxG1, while maintaining NCS self-renewal. Bmi-1 overexpression restored the maintenance of NSCs in vitro. FoxG1 was found to promote the methylation of Foxo3 promoter and inhibited Foxo3 expression. miR-186 upregulation increased the expression of Foxo3 and inhibited NSC self-renewal in the absence of Foxo3. Therefore, we propose that downregulation of miR-186 maintained NSC self-renewal in the postnatal brain by upregulat...

Roles of Immune and Oxidative Stress-Related Factors in the Diagnosis of Coronary Artery Disease: A Retrospective Study
CONCLUSIONS: Bioinformatics analysis technology was used to identify 10 hub DEIOGs that might play a crucial role in CAD progression, and five special hub genes (Fos, Il6, Jun, Mapk3, and Mmp9) could be regarded as potential biomarkers for CAD diagnosis. However, further studies are required to verify the functions of these hub genes.PMID:37691276 | DOI:10.59958/hsf.5799

Molecular complexity of mammary glands development: a review of lactogenic differentiation in epithelial cells
Artif Cells Nanomed Biotechnol. 2023 Dec;51(1):491-508. doi: 10.1080/21691401.2023.2252872.ABSTRACTThe mammary gland is a dynamic organ with various physiological processes like cellular proliferation, differentiation, and apoptosis during the pregnancy-lactation-involution cycle. It is essential to understand the molecular changes during the lactogenic differentiation of mammary epithelial cells (MECs, the milk-synthesizing cells). The MECs are organized as luminal milk-secreting cells and basal myoepithelial cells (responsible for milk ejection by contraction) that form the alveoli. The branching morphogenesis and lactog...

Colorectal cancer cell-secreted exosomal miRNA N-72 promotes tumor angiogenesis by targeting CLDN18
Am J Cancer Res. 2023 Aug 15;13(8):3482-3499. eCollection 2023.ABSTRACTAngiogenesis is essential for the growth and metastasis of several malignant tumors including colorectal cancer (CRC). The molecular mechanism underlying CRC angiogenesis has not been fully elucidated. Emerging evidence indicates that secreted microRNAs (miRNAs) may mediate the intercellular communication between tumor cells and neighboring endothelial cells to regulate tumor angiogenesis. In addition, exosomes have been shown to carry and deliver miRNAs to regulate angiogenesis. miRNA N-72 is a novel miRNA that plays a regulatory role in the EGF-induce...

CircCRIM1 suppresses osteosarcoma progression via sponging miR146a-5p and targeting NUMB
Am J Cancer Res. 2023 Aug 15;13(8):3463-3481. eCollection 2023.ABSTRACTCircCRIM1 (hsa_circ_0002346) is a circular RNA derived from gene CRIM1 (the cysteine rich transmembrane BMP regulator 1 circRNAs) by back-splicing. Recent studies have suggested the diverse function of CircCRIM1 in the tumorigenesis of multiple malignancies, including osteosarcoma (OS). Here, we investigated the role and mechanism of circCRIM1 during OS progression. Differentially expressed circRNAs (including circCRIM1) in OS and human osteoblast (hFOB1.19) cell lines were selected by searching the circRNA expression microarray dataset of GSE96964. The...
